Objective:1.Using data mining methods,we summarize and analyze the drug use of traditional Chinese medicine in the therapy of spleen deficiency,phlegm and dampness type PCOS characteristics and patterns,and uncover core drug combinations and potential new prescriptions.To provide support for the document research and clinical utilization of traditional Chinese medicine in the therapy of PCOS with spleen deficiency,phlegm and dampness,and to provide directions for new drug development.2.To predict and explore the potential target pathways and their mechanisms of action of core prescriptions for the therapy of spleen deficiency,phlegm and dampness type PCOS using a network pharmacology approach.To explore more avenues for the herbal treatment of spleen deficiency,phlegm and dampness type PCOS and to lay the theoretical basis for subsequent experimental studies.Method:1.DATA MINING:CNKI,WANGFANG DATA,VIP and CBM databases were used to retrieve the literature on traditional Chinese medicine for spleen deficiency,phlegm and dampness type PCOS since the establishment of the database to 09,2022.The database was established by EXCEL form,and the data of literature name,first author,disease diagnosis,literature type,journal name/author unit,drug composition and dosage,and literature publication date were counted.With the help of TCMISS(V2.5)system,basic information,formula pattern and new formula combinations were analyzed,and highfrequency drug combinations for the treatment of spleen deficiency,phlegm and dampness type PCOS were mined.2.Network Pharmacology:The core drug combinations with support≧50 were used as core prescriptions,and the active ingredients and targets were screened by TCMSP database.TCMSP database for active ingredient and target screening;Uniprot database for target specification;Cytoscape 3.9.1 software for "core prescription-active ingredient-target" visualization network diagram.The target screening was performed in DrugB ank,OMIM,GeneCard,PharmGKB and DisGeNET databases;the intersection of "core prescriptiondisease" was mapped using the Venny function of OmicShare software.PPI network construction of intersection targets by String database,extraction of Hub-Target based on MCC algorithm by Cytoscape3.9.1 CytoHubba plug-in,GO biofunctional analysis and KEGG signaling pathway analysis by DAVID6.8 database,and the use of Cytoscape 3.9.1 software was used to map the "core prescription-active ingredient-target-disease-pathway" network to predict the core targets of the core prescription for spleen deficiency and phlegm.The core targets,pathways and potential mechanisms of action of the core prescription for the treatment of spleen-deficient phlegm-damp PCOS were predicted.Result:1.DATA MINING:(1)Analysis of basic information:92 articles with 102 prescriptions were finally included in this study.A total of 167 traditional Chinese medicine were included,and the top 10 traditional Chinese medicines in order of frequency were:Fu Ling(82 times),Chen Pi(66 times),Radix Aromaticus(62 times),Atractylodes Macrocephalae(62 times),Radix Panax(60 times),Atractylodes Macrocephalae(58 times),Radix Angelicae Sinensis(55 times),Radix Glycyrrhiza Uralensis(48 times),Cuscutae(38 times),and Rhizoma Chuanxiong(37 times).The four natures of drugs are mainly warm,the five flavours are mainly sweet,and the meridians are mainly the spleen,liver and kidney meridians.(2)Analysis of formula pattern:There are 46 drug combinations with frequency≧30 times,including Chen Pi,Poria,Semen,Atractylodes,Atractylodes,Angelica,Glycyrrhiza and Chuanxiong,Poria,Radix et Rhizoma,Atractylodes Macrocephala,Rhizoma Atractylodes,Rhizoma Atractylodes Macrocephala,Radix Angelicae Sinensis,Radix et Rhizoma Glycyrrhizae,Rhizoma Ligustici Chuanxiong and other 9 traditional Chinese medicine.The core prescription drugs combination with a support degree of≧50 was "Fu Ling,Chen Pi,Xiang Fu,Atractylodes Macrocephala,and Radix Panax notoginseng".Using the core drug group as the core prescription,the mechanism of action was further analyzed based on the network pharmacology method.(3)New formula analysis:Based on the improved mutual information method,the drug pair correlation analysis,in which the highest correlation The drug pair with the highest degree of association was "Cuscuta sinensisraspberry".Based on the entropy clustering of complex systems,20 sets of potential core combinations were co-clustered,such as "Angelica sinensisHawthorn-Lotus leaf",etc.Based on unsupervised entropy hierarchical clustering of new prescriptions,six potential new prescriptions for the treatment of spleen deficiency,phlegm and dampness type PCOS were clustered,such as "bamboo rhizome-turtle nail-heliotrope leaf-gingerdeer horn gum "etc.2.Network Pharmacology:(1)Core prescription active ingredients and target screening:44 active ingredients in the core prescription;256 potential targets.According to the analysis of "core prescription-active ingredient-target" diagram,the main active component of the core prescription involved quercetin,β-sitosterol,dousterol,kaempferol and lignocaine.There are 1957 disease targets in polycystic ovary syndrome.There were 126 core prescription and disease intersection targets.(2)After topological analysis,the core targets of this study were AKT1,TP53,MAPK3,MAPK1,IL6,TNF,FOS,ESR1,EGFR,RB1.These target proteins were shown to play an important role in the treatment of spleen deficiency and sputum wet PCOS.(3)GO biofunctional and KEGG signaling pathway enrichment analyses were performed on the intersection targets.The results of GO enrichment analysis showed that BP mainly includes drug response,positive regulation of gene expression,positive regulation of cell proliferation,negative regulation of the apoptotic process,etc.CC mainly includes protoplasmic membrane,extracellular fluid,extracellular region,integral part of plasma membrane,etc.MF mainly includes protein binding,identical protein binding,enzyme binding,cytokine activity,etc.The results of KEGG enrichment analysis predicted that the core prescription may be exerted by regulating cancer,PI3KAkt,lipid and atherosclerosis,and diabetic complications AGE-RAGE signaling pathways for the treatment of spleen-deficient phlegm-damp PCOS.Conclusion:1.In this study,the full text search and summary of clinical literature on the treatment of PCOS with spleen deficiency,phlegm-dampness type with traditional Chinese medicine were carried out,and the results of TCMISS analysis showed that the high-frequency drug combinations in the literature on the treatment of PCOS with spleen deficiency,phlegm-dampness type were"Fu Ling,Chen Pi,Xiang Fu,Atractylodes,and Sempervivum ".Traditional Chinese medicine is mainly warm and calming drugs,with sweet taste as the main medicine and spleen channel.2.The treatment of PCOS of spleen deficiency and phlegm-dampness with traditional Chinese medicine mainly focuses on eliminating phlegm and dampness and regulating collaterals and meridians.Based on the entropy cluster analysis of complex system,six new prescription combinations were obtained,such as "Zhuru-turtle shell-Phoschinia leaf-ginger-deer horn gum",etc.,which opened up ideas and directions for the clinical prescription use of spleen deficiency,phlegm-dampness type PCOS and the research and development of new drugs.Its efficacy needs to be further verified in clinical practice.3.Network pharmacology predicts that the core prescription(Fu Ling,Chen Pi,Xiang Fu,Atractylodes,and Sempervivum)may act on targets such as AKT1,TP53,MAPK3,MAPK1 and IL6,through active active ingredients such as quercetin,β-sitosterol,dousterol,kaempferol,and lignocaine,which are involved in cancer,PI3K-Akt,lipid and atherosclerosis,diabetic complications AGE-RAGE and other signaling pathways to play a role in the treatment of spleen deficiency,phlegm and dampness type PCOS.Reflecting the characteristics of multi-component,multi-target and multi-pathway of herbal compound,the three synergistic effects for the treatment of spleen deficiency phlegm and dampness type PCOS. |
