Objectives To observe the repair effect of Jingui Wenjing decoction on endometrial injury in a rat model of gynecological deficiency cold syndrome.Methods A total of 50 female SPF-grade SD rats aged 3 months and weighting 180-220g were selected.The rats were randomly divided into normal group,model group,low-dose Jingui Wenjing decoction group,medium-dose Jingui Wenjing decoction group and high-dose Jingui Wenjing decoction group,with 10 rats in each group.Except for the normal group,a model of gynecological deficiency cold syndrome was established in the other groups by freezing in a refrigerator+ice water immersion.The low-dose Jingui Wenjing decoction group,medium-dose Jingui Wenjing decoction group and high-dose Jingui Wenjing decoction group were given gavage administration with 4.7 g/kg/d,9.4g/kg/d and 18.8 g/kg/d Jingui Wenjing decoction every day,respectively.The normal group and the model group were given gavage administration with equal volume of distilled water.The general condition of the rats was observed,and the estrous cycle of the rats were observed using vaginal smears.Thirty days after establishment,the rats were sacrificed at the estrous interval,and then the serum and uteruses were collected.The contents of E2,T and P in the serum were detected using ELISA.The pathological morphology of the uteruses was observed by HE staining,and the thickness of the endometrium was calculated.The protein localization of ER,PR,CD31 andα-SMA in the uterine tissue was determined by immunohistochemical staining.The protein expressions of JAK1,LIF,LIFR,phosphorylated-signal transducers and activators of p-STAT3 and VEGF in the uterine tissue were detected by Western blotting.Results 1 Compared with the normal group,the rats in the model group were in low spirits,curled up and moved less,and their fur color was not bright,fingernails,ears and tongues were dark purple,stools were loose,intake of food and water decreased,and body weight increased slowly.E2,P and T contents in the serum reduced.HE staining showed endometrial thinning in the uterine tissue.Immunohistochemistry presented that the expressions of ER,PR,CD31 andα-SMA reduced.Western blotting revealed that the expressions of JAK1,LIF,LIFR,p-STAT3 and VEGF decreased.2 Compared with the model group,the rats in the low-dose,medium-dose and high-dose Jingui Wenjing decoction groups were in normal spirits and moved more,and their fingernails,ears and tongues were light red,stools were not loose,intake of food and water increased slightly,and body weight increased rapidly.E2,P and T contents in the serum increased.The endometrial thickness increased.Immunohistochemistry revealed that the expressions of ER,PR,CD31 andα-SMA increased.Western blotting showed that the expressions of JAK1,LIF,LIFR,p-STAT3 and VEGF increased.Conclusions 1 Endometrial injury occurs in rat models of gynecological deficiency cold syndrome.2 Jingui Wenjing decoction can regulate the expression of hormone receptors and blood vessel-related factors in the endometrium and repair the injured endometrium of rats with gynecological deficiency cold syndrome.Figure 6;Table 6;Reference 95...
