Stellera chamaejasme L.is a perennial herb belonging to the Stellera genus of the Thymelaeaceae family.Its dry roots called“langdu”are first recorded in Shennong’s Classic of Materia Medica,which have long been used in traditional Chinese to treat stubborn skin-related diseases,chronic tracheitis and tuberculosis.Modern pharmacodynamic components and pharmacological studies show that the principal chemical constituents include sesquiterpenoids,diterpenoids,flavonoids,coumarins.Among them,terpenoids show excellent biological activities in antitumor,antiviral,antibacterial and anti-inflammatory aspects.In order to further reveal the antitumor pharmacodynamic substance basis,compounds of the roots of S.chamaejasme were systematically isolated and identified,and the sesquiterpenoids were tested for antitumor and antibacterial activities.In this study,42 compounds were isolated and identified by using a variety of column chromatography preparation methods and modern spectroscopic instruments.These compounds include 16 sesquiterpenoids:chamaejasmone D(A1),daphnauranol B(A2),stelleraguaianone B(A3),holosericin B(A4),chamaejasmin C(A5),chamaejasmin B(A6),auranticanol A(A7),1α,7α,10αH-guaia-4,11-dien-3-one(A8),chamaejasmin A(A9),daphnauranol D(A10),stelleraterpenoid B(A11),chamaejasnoid A(A12),chamaejasnoid B(A13),chamaejasnoid C(A14),chamaejasnoid D(A15)and chamaejasnoid E(A16);3 lignans:dextrobursehernin(A17),hinokinin(A18)and matairesinol(A19);9 flavonoids:naringenin(A20),ruixianglangdusu B(A21),chamaejasmenin B(A22),isochamaejasmin(A23),chamaejasmenin A(A24),(-)-afzelechin(A25),chamaechromone(A26),neochamaejasmin A(A27)and neochamaejasmin B(A28);3 coumarins:daphnoretin(A29),umbelliferone(A30)and 7-hydroxy-8-methoxycoumarin(A31);3 steroids:(24R)-24-propylcholesterol(A32),β-sitosterol(A33)andβ-sitosteryl acetate(A34);and other typle compounds:daphnecinnamte A(A35),glyceryl monopalmitate(A36),(+)-3-hydroxy-1,5-diphenyl-l-pentanone(A37),1,5-diphenyl-2-penten-1-one(A38),trans-cinnamic acid(A39),1-octacosanol(A40),p-hydroxybenzoic acid(A41)and pyrocatechol(A42).Among them,chamaejasnoid A(A12)is an unprecedented 2,3-seco-guaiane sesquiterpenoid with a 5/6/7bridged ring,in particular harboring an rigid 3,6-dioxabicyclo[3.2.1]octane-4,7-dione moiety,and chamaejasnoid B(A13)is a unique 2-nor-guaiane sesquiterpenoid.Some sesquiterpenoids were tested for cytotoxicity,among which A16 had strong killing effect on human colon cancer cells(HCT8),and its IC50 was 11.82±2.89μM.
