ObjectiveThe expression levels of EZH2(enhancer of zeste homolog 2),EED(embryonic ectoderm development)and SUZ12(suppressor of zeste 12),the core components of the PRC2 protein(Polycombrepressive complex 2),were measured in the chorionic villi of patients with missed abortion and normal pregnancy abortion,and to investigate the effect and mechanism of these three ingredients in abortion.Thus,the theory foundation can be set up to recognize,intervene and treat the missed abortion in time.Methods1.30 patients diagnosed with missed abortion who attended or were hospitalized at the Third People’s Hospital of Hefei from January 1,2022 to December 31,2022 were selected as the experimental group of missed abortion(MA group),and 30 patients with normal pregnancy who voluntarily requested for painless abortion were the control group of normal miscarriage.2.The sections of both groups were treated with hematoxylin-eosin staining(HE,hematoxylin-eosin staining)and the histological changes of the chorionic villus tissue in both groups were observed under a light microscope.The expression levels and differences of EZH2,EED and SUZ12 in the chorionic villus tissues of the two groups were determined using protein blotting(WB,western blot).The distribution and differences of EZH2,EED and SUZ12 in the chorionic villus tissues of the two groups of patients were observed by immunohistochemistry(IHC Immunohistochemistry).Results1.Comparison of general conditions of patients in the control group with induced miscarriage and the experimental group with missed abortion(MA group)There was no statistical difference between the control group and the experimental group in terms of age,gestation,number of births and gestational weeks(P>0.05).2.Histological differences between the control group with induced miscarriage and the experimental group with missed abortion(MA group)in terms of HE staining of chorionic villus tissueAfter HE staining,the sections were examined under a light microscope and showed that in the induced abortion control group: the chorionic villous tissue was regular in shape,clearly layered and structurally intact,and the trophoblast cells were uniform in size,regular in shape,with clear borders and neatly arranged;in the experimental group with missed abortion:some of the villi are morphologically altered,with poorly layered,disorganized and damaged structures,trophoblast cells of varying sizes,reduced in number,irregular,poorly defined borders,disorganized arrangements,visible nuclear consolidation,and interstitial villi showing varying degrees of oedema and fibrous degeneration.3.The expression levels of EZH2,EED and SUZ12 in the villi of the control group with induced abortion and the experimental group with missed abortion(MA group)The results of protein blotting showed that the expression levels of EZH2,EED and SUZ12 in the chorionic villus tissues of the experimental group with missed abortion were significantly lower than those of the control group and the difference between the both groups is remarkable(P<0.05).The immunohistochemical results showed that the expression levels of EZH2,EED and SUZ12 in the chorionic villus tissues of the induced abortion control group were higher than those of the missed abortion experimental group,and the change between the two groups was considered highly(P<0.05).ConclusionThe findings show that the expression levels of EZH2,EED,and SUZ12 are downregulated in the chorionic tissue of patients with missed abortion,and because gene expression is essential for embryonic development and the PRC2 protein serves as a key chromatin modification complex,the results of this experiment highlight the possible regulatory role of the PRC2 protein in embryonic development,and the absence of PRC2 protein may contribute to the development and progression of missed abortion.This provides part of the rationale for proactive preventive interventions of patients with missed abortion in clinical practice. |
