| Objective: Metabolomics technology was used to search for potential biomarkers of Qufengjuanyuan decoction in the treatment of cold asthma in children,and to explore the pathogenesis of asthma and the mechanism of action of Qufengjuanyuan decoction in the treatment of cold asthma in children.Research methods: A total of 26 children with cold asthma were selected from the Department of Traditional Chinese Medicine of Shenzhen Children’s Hospital from September 2020 to September 2021,and were treated with Qufengjuanyuan decoction,and 30 healthy children were selected from the Department of Paediatrcare.Urine samples of children with asthma were collected before treatment、after treatment and healthy control group,respectively.Liquid chromatography tandem mass spectrometry(LC-MS/MS)was used to detect urine samples of children with asthma Qufengjuanyin Decoction before and after treatment and healthy control group.Principal component analysis was used(PCA)and Partial least squares discriminant analysis(PLS-DA)to search for the differential metabolites of bronchial asthma with cold symptom and Qufengjuanyin decoction in the treatment of asthma.The metabolic changes in children with asthma and those before and after treatment with Qufengjuanyin Decoction were analyzed,and the pathogenesis of asthma and the action mechanism of Qufengjuanyin decoction in the treatment of asthma were preliminarily explored.Results: 1.A total of 56 subjects aged from 5 to 14 years old were included in this study,including 30 healthy control patients and 26 children with bronchial asthma with cold symptom.There were no statistical differences in gender and age of children(P > 0.05).A total of 81 urine samples were collected in this study,including 30 healthy control cases,26 cases before treatment with Qufeng juanyin Decoction and 25 cases after treatment with Qufeng juanyin decoction in children with cold asthma.2.The LC-MS/MS technique was used to detect the urine samples of children with bronchial asthma and healthy children.Combined with PCA and PLS-DA,it was found that the urine metabolic profile of healthy children and children with bronchial asthma with cold symptom was different.And eventually found Anserine,DCA,GDCA,HArg,11-Met-His,Acon Acid,Cys,DOPA,Dopamine,Hypoxanthine,PC aa C38:6,Phe Ala Betaine,Putrescine,SDMA,Serotonin etc 20 different metabolites.3.LC-MS/MS technology was used to detect urine samples of children before and after the treatment of bronchial asthma with Qufengjuanyan Decoction.Combining PCA and PLS-DA,differences were found in urine metabolic profile of children with bronchial asthma with Qufengjuanyan decoction before and after the treatment.And finally found1-Met-His,3-IAA,ADMA,Asp,Betaine,C3-DC Dopamine(C4-OH),CA,Cystine,DOPA,GABA,Glu,Histamine,Hypoxanthine,Ile,Met-SO,PEA,SDMA,Serotonin,Spermidine,TG(17:0_34:1),etc 30 differential metabolites.4.The quantitative results of the two groups were analyzed,and it was found that Qufengjuanyin decoction restored the levels of the following 9 biomarkers: 1-Met-His,DOPA,Hypoxanthine,Pro Betaine,Putrescine,Serotonin,SDMA,TG(17:0_34:1),DopamineConclusions: 1.Through the analysis of 20 different metabolites in urine of healthy children and children with cold asthma,it was found that the abnormal metabolism of amino acids,bioamines and lipids in children with cold asthma was closely related to the mechanisms of energy metabolism,oxidative stress and inflammation in the body.2.Through the analysis of 30 different metabolites in urine of children with bronchial asthma with cold asthma before and after treatment with Qufengjuanyin Decoction,it was found that the treatment with Qufengjuanyin decoction could improve the metabolism of amino acids,bioamines and lipid in the children,and improve airway inflammation in children with cold asthma by improving oxidative stress and inflammation.Relieve the clinical symptoms of children with bronchial asthma with cold asthma.3.By comparing the levels of metabolites before and after treatment with healthy children and children with cold asthma syndrome of bronchial asthma and children with cold asthma syndrome of bronchial asthma,it is found that Qufengjuanyin decoction may improve oxidative stress and inflammatory response by affecting 9 metabolites that are regulated,so as to achieve the purpose of treating cold asthma syndrome of children. |
