Objective: To establish the fingerprint of Rumex japonicus Houtt.(R japonicus),a traditional Chinese medicine,to ensure the quality consistency of the medicinal materials;To study the antifungal material basis of this medicinal material based on the antifungal activity of R japonicus antibody;to establish a network pharmacological targeting pathway for the treatment of dermatophytic related fungi with this medicinal material;To verify the antifungal activity of this medicinal material against dermatophytes,and to conduct preliminary mechanism studies.Methods: The fingerprints of five-year-old R japonicus from different regions were established by high performance liquid chromatography(HPLC),and evaluated and analyzed by methods such as stoichiometry;the chemical components of the R japonicus was studied by comprehensive chromatography and nuclear magnetic resonance(NMR)technology;Using network databases and software such as R and Cytascape to conduct network pharmacological research;Microdilution method,extracellular protein assay,and XTT reduction method were used to study the anti dermatophytic fungi activity in vitro.Results: the study on the fingerprint of the five-year-old R japonicus from different regions was completed,the 18 common peaks were identified with the 10 characteristic peaks for the first time.The structures of 11 chemical components from the R japonicus were elucidated and a systematic and comprehensive structural assignment of them was carried out,including Rhein,Chrysophanol,Emodin,Emodin methyl ether,Aloe emodin,Rutin,Quercetin,and Chrysophanol-8-O-β-D-glucopyranoside,Emodin-8-O-β-D-glucopyranoside,Ferulic acid,and p-Coumarin acid.a network pharmacology related targeting pathway has been established between the R japonicus and the common fungi like Trichophyton trichophyton,Trichophyton rubrum,Microsporium canis,and Candida albicans,as well as the antifungal activity of different chemical fractions and pure compounds of the R.japonicus.The results showed that the R japonicus mainly had strong antifungal activity against Trichophyton trichophyton,and the related mechanism was studied also.Conclusion: The establishment of a system related to the activity of the five-year-old R japonicus fingerprint spectra,ensuring the consistency of the medicinal application and quality evaluation of R japonicus;The chemical structures of the isolated chemical components were studied to improve their structural attribution,laying an experimental foundation for further research on the chemistry of anthraquinones and flavonoids in the R japonicus;Network pharmacology provides a targeted pathway for studying the pharmacology and related mechanisms of anti dermatophytic fungi;the studies on the activity and mechanism of anti dermatophytic fungi have identified their targeted chemical fractions and monomer compounds,providing a basis for further drug development,and laying an experimental foundation for the rational design of safe and effective new drugs.
