Objective: To summarize the commonly used prescriptions,major Chinese medicines and medication rules of Professor Dexi Zhao in the treatment of ischemic stroke.Further,to explore the preliminary mechanisms of commonly used prescriptions in the treatment of ischemic stroke.Methods: 1.Patients treated by Professor Dexi Zhao at the Affiliated Hospital of Changchun University of Traditional Chinese Medicine from January 2018 to December2022 that conform to inclusion and exclusion criteria were selected.The basic information of the medical records was analyzed by applying the TCM inheritance assistance platform V3.0.These analyses include major herbs and commonly used prescriptions and among others for the treatment of ischemic stroke by Professor Dexi Zhao.2.The active components of the commonly used prescription(Dachaihu decoction)were screening from HPLC literature,and the corresponding active components’ targets were obtaining from Swiss Target Prediction,HERB,ETCM and Super-PRED databases.Targets of ischemic stroke were obtained from datasets GSE58294 and GSE66724 in GEO database and Gene Cards and OMIM databases.Potential targets of Dachaihu decoction for ischemic stroke obtained from Venny2.1.Then,the components-targets network was constructed using Cytoscape software and screening key targets.The GO and KEGG enrichment analysis of the potential targets were performed by Metascape.Finally,molecular docking of major components and targets was performed.Results: 1.200 patients who conform to inclusion and exclusion criteria were included in this study.Of the 200 patients,107(53.5%)were men and 93(46.5%)were women.The highest prevalence was in the age range of 51-60 years.2.The most common patients included were syndrome of Feng Tan Zu Luo and Feng Huo Shang Rao.The the commonly used prescriptionst for syndrome of Feng Tan Zu Luo was used Ban Xia Bai Zhu Tianma decoction and Wen Dan decoction.And the commonly used prescriptions for syndrome of Feng Huo Shang Rao was used Dachaihu decoction.3.Professor Dexi Zhao’s treatment of ischemic stroke is based on the treatment of invigorating spleen for diuresis,promoting blood circulation for removing blood stasis,and reconciliation method,with deficiency tonics as the mainstay of treatment,followed by blood activators and heat purifiers.The four gases of the herbs are mainly cold,warm and flat,and the five tastes are mainly pungent,sweet and bitter.The main channels of drugs are liver,spleen,stomach and lung,etc.The major herbs for the treatment of ischemic stroke are Rhizoma Pinelliae,Radix Bupleuri,Fructus Zizyphi Jujubae,Rhizoma Zingiberis Recens,Fructus Aurantii Immaturus,Poria,Eupolyphaga seu Steleophaga,Hirudo,Glycyrrhizae Radix et Rhizoma and among others.4.The active components in the commonly used prescription Dachaihu decoction are emodin,paeoniflorin and baicalin,etc.which have anti-inflammatory and antioxidant functions and regulation of various signaling pathways.AKT1,MMP9 and IL1 B are the key targets for the treatment of ischemic stroke,and PI3K-AKT,MAPK,cGMP-PKG signaling pathway and apoptosis may be the main mechanisms of Dachaihu decoction for the treatment of ischemic stroke.The molecular docking of core targets and major active components has high docking score.Conclusion: Based on data mining and network pharmacology,this study explores the medication rules of Professor Dexi Zhao’s treatment of ischemic stroke and the potential mechanism of commonly used prescription Dachaihu decoction in the treatment of ischemic stroke,in order to provide ideas for the treatment of ischemic stroke and provide a theoretical basis for further experimental studies.
