Objective: To evaluate the clinical efficacy of Yinqiao Ma Bo powder in the treatment of laryngeal cough caused by moisture and heat accumulation and lung qi nondeclaration,observe the scores of symptoms and signs before and after treatment by self-comparison method,so as to compare and confirm the clinical efficacy of this prescription,and further expand the clinical reference significance of Nemesis in the treatment of laryngeal cough.At the same time,network pharmacology was used to dig out the active ingredients,action targets and corresponding pathways of Yinqiao Ma Bosan in the treatment of laryngeal cough,and to analyze and find its potential mechanism of action,providing the basis for the preliminary research of Yinqiao Ma Bosan in the treatment of laryngeal cough.Methods: From July 2021 to July 2022,60 follow-up cases of laryngeal cough meeting the diagnostic criteria were included,and the symptoms and signs before and after medication were recorded and scored.The scoring data were analyzed and verified by self-before-and-after control method.At the same time,the online TCMSP database platform was used to search,so as to obtain the active ingredients and targets of drugs,and the obtained targets were genetically processed using the online Uni Prot database.In this study,the online Gene Cards gene bank platform was used to search for "geal cough" in laryngeal cough.Venny 2.1.0 online tool was used to analyze and obtain the common targets of diseases and drugs.Active components and common targets were input into Cytoscape3.8.0 to generate the composition-target network diagram.The protein network interaction diagram was obtained by online STRING database,and the core targets were screened out.Finally,GO biological process analysis and KEGG pathway enrichment analysis of core targets were conducted using online Metascape database.Results: According to statistical analysis,in the treatment of laryngeal cough with Yinqiao Mabbo powder,the biggest symptom relief was sore throat(76.4%)and foreign body sensation in pharynx(72.7%).According to the total effective rate,cough was the highest(96.7%).The effective rate of other symptoms was > 90%,and in descending order were bitter mouth(93.8%),pharyngeal congestion,follicular hyperplasia of posterior pharyngeal wall(92.9%),and pharyngeal pain(91.7%).90% >effective rate > 80%,in order of high to low,they were respectively pharyngeal itch(86.4%),chest tightness(85.7%),foreign body sensation in pharynx(82.4%).80% > effective rate > 70%,from high to low were sputum(80.0%),dry throat(70.6%).The scores of symptoms and signs before and after were significantly different(p < 0.01).A total of 33 effective components of Yinqiao Ma Bo powder for laryngeal cough were selected by network pharmacology.Among them,luteolin,quercetin,kaempferol,stigmatosterol,baicalin and β-carotene may be the key components in the treatment process.It involved 26 core targets such as AKT1,TP53,TNF,ESR1,MYC,EGFR,IL6 and CAV1.These targets may be attached to TNF signaling pathway,C-type lectin receptor signaling pathway,RIG-I receptor signaling pathway,Toll-like receptor signaling pathway,No D-like receptor signaling pathway,IL-17 signaling pathway and other pathways.Conclusion: This study suggests that Yinqiao Ma Bosan,as the main prescription,can significantly improve the symptoms of cough,pharyngeal pain,pharyngeal itch,dry pharyngeal,foreign body sensation in pharyngeal,bitter mouth and other symptoms in patients with laryngeal cough.At the same time,it can also significantly improve the signs of follicular hyperplasia and congestion in the posterior pharyngeal wall,without obvious adverse reactions,and has definite clinical efficacy.Yinqiaoma Bossan is used to treat laryngeal cough by acting on multiple targets such as AKT1,TP53,TNF,ESR1 and multiple signaling pathways such as TNF,RIG-I receptor and toll-like receptor.It not only validates the scientific significance of the compatibility of traditional Chinese medicine with the king,minister,complator and agent.At the same time,it provides some meaningful theoretical data for basic research.
