| Objective: To observe the expression of serum interleukin in Henoch Schonlein purpura(HSP),Henoch Schonlein purpura nephritis(HSPN)and healthy children with heat toxin and blood stasis syndrome,and to explore the expression of IL-2,IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α In the pathogenesis of Henoch Schonlein purpura and Henoch Schonlein purpura nephritis,and the efficacy and mechanism of detoxification and stasis removal in the treatment of Henoch Schonlein purpura and Henoch Schonlein purpura nephritis.Materials and methods: All cases were from the outpatient department of the Affiliated Hospital of Liaoning University of traditional Chinese medicine.16 adolescents aged 5-18 years who dialectically diagnosed with Henoch Schonlein purpura with heat toxin and blood stasis syndrome were selected as the purpura group(HSP group),the HSP group after treatment with Zidan decoction,and 10 normal adolescents were selected as the healthy group(H Group).The expression of various cytokines was detected by Luminex detection technology,and the obtained data were imported into SPSS 23.0 software for relevant statistical analysis.Result:1.In HSP group,the serum IL-2 level was 6.99 ± 2.14pg/ml,the IL-5 level was 3.89 ±2.41pg/ml,the IL-6 level was 42.66 ± 26.92pg/ml,the IL-10 level was 6.96 ± 1.40pg/ml,the G-CSF level was 53.45 ± 28.67pg/ml,the MCP-1 level was 207.49 ± 30.70pg/ml,TNF-α The level was 12.95 ± 5.04pg/ml;In the healthy group,the serum IL-2 level was14.01 ± 2.42pg/ml,IL-5 level was 1.14 ± 0.48pg/ml,IL-6 level was 6.65 ± 5.20pg/ml,IL-10 level was 2.33 ± 0.86pg/ml,G-CSF level was 10.66 ± 4.58pg/ml,MCP-1 level was132.59 ± 12.79pg/ml,TNF-α The level was 6.84 ± 2.23pg/ml.In the comparison between HSP group and healthy group,serum IL-2,IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α The levels were significantly different(all P < 0.05).2.In HSP group,serum IL-2 level was 12.50 ± 2.58pg/ml,IL-5 level was 1.34 ±0.68pg/ml,IL-6 level was 7.72 ± 7.46pg/ml,IL-10 level was 2.90 ± 1.21pg/ml,G-CSF level was 14.59 ± 8.06pg/ml,MCP-1 level was 148.50 ± 27.54pg/ml,TNF-α The level was 6.95 ± 2.17pg/ml.In the comparison of HSP group before and after treatment,serum IL-2,IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α There were significant differences in levels(all P < 0.05);After treatment,compared with the healthy group,serum IL-2,IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α There was no significant difference in level(P > 0.05).3.According to the correlation analysis of serum IL-2,IL-5,IL-6 and IL-10 in children with HSP before treatment,there was no correlation between the expression level of IL-2 and IL-5(R2 = 0.005,P = 0.98 > 0.05);The expression levels of IL-2 and IL-6 were negatively correlated(R2 = 0.29,P < 0.05);The expression levels of IL-2 and IL-10 were negatively correlated(R2 = 0.59,P < 0.05);There was no correlation between the expression levels of IL-5 and IL-6(R2 = 0.04,P = 0.45 > 0.05);There was no correlation between the expression level of IL-5 and IL-10(R2 = 0.02,P = 0.6 > 0.05);There was a positive correlation between the expression level of IL-6 and IL-10(r = 0.35,P < 0.05).4.After 4 weeks of treatment,the cure rate of children with Henoch Schonlein purpura was87.5%.Conclusion:1.Before treatment,the level of IL-2 in serum of Henoch Schonlein purpura group decreased,including IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α These factors may be involved in the pathogenesis of Henoch Schonlein purpura.2.After traditional Chinese medicine treatment,the cytokine level changes and returns to normal,suggesting that traditional Chinese medicine intervention is effective in the treatment of Henoch Schonlein purpura.3.The treatment of Henoch Schonlein purpura by detoxifying and removing blood stasis can significantly reduce the clinical symptoms and signs of children.Its mechanism may be related to inhibiting the synthesis and secretion of inflammatory cytokines,inhibiting the inflammatory response and enhancing the effect of anti-inflammatory factors.Therefore,it is speculated that IL-2,IL-5,IL-6,IL-10,G-CSF,MCP-1 and TNF-α It provides a powerful basis for the use of Zidan Decoction in the treatment of Henoch Schonlein purpura. |
