Objective: To investigate the effect and mechanism of Bushen Huoxue decoction on bone metabolism in ovariectomized osteoporosis mice by regulating PI3K/AKT/mTOR pathway.Methods: A mouse model of postmenopausal osteoporosis was established by OVX method.Forty SD female mice were randomly divided into sham operation group,model group,western medicine(estradiol valerate suspension)group and Bushen Huoxue decoction group,with 10 mice in each group.After successful modeling,the mice in the sham operation group and the model group were given the same amount of normal saline by gavage,the mice in the western medicine group were given estradiol valerate and normal saline suspension by gavage,and the mice in the Bushen Huoxue decoction group were given the same dose of Bushen Huoxue decoction by gavage.After 6 weeks of continuous administration,the femoral bone mineral density of mice was detected by dual-energy X-ray,the pathological changes of femur tissue were observed by HE staining,the serum BALP and TRAP levels were detected,and the protein expressions of PI3 K,AKT and mTOR in the femur of mice were detected by Western blot.Results:(1)Compared with the sham-operation group,the bone mineral density of the model group decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the BMD of the western medicine group and the Bushen Huoxue decoction group increased,and the difference was statistically significant(P<0.05).(2)HE staining of mouse femur: in the sham operation group,the bone cortex and cancellous parts were intact,the trabecular structure was tight,and the matrix staining was uniform.In the model group,the defects of bone cortex and cancellous bone,the number of bone trabeculae decreased,the gap increased,the continuity was interrupted,the structure was sparse,the bone marrow cavity became wider,the bone cortex became thinner,and the matrix staining was severely reduced,which was consistent with the pathological changes of osteoporosis.In the western medicine group,the bone cortex and cancellous bone were slightly defective,the matrix staining was slightly reduced,the number of bone trabeculae was increased compared with the model group,the structure was improved,and the continuity was good.The bone cortex was basically similar to the sham-operation group.The bone tissue staining in the Bushen Huoxue decoction group was roughly the same as that in the western medicine group.(3)Compared with the sham-operation group,the serum BALP level of the model group was decreased,and the TRAP level was increased(P<0.05).Compared with the model group,the serum BALP level of the western medicine group and the Bushen Huoxue decoction group increased,and the TRAP level decreased,and the difference was statistically significant(P<0.05).(4)Compared with the sham operation group,the expression of PI3 K,AKT and mTOR in the model group decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of PI3 K,AKT and mTOR in the intervention group was increased,and the difference was statistically significant(P<0.05).Conclusion: Bushen Huoxue decoction can reduce bone loss and inhibit bone resorption in OVX mice model of osteoporosis,and its effect may be related to the activation of PI3K/AKT/mTOR signaling pathway.
