Objective Through animal experiments,the effect of Bushen Huoxue Decoction on glucocorticoid-induced osteoporosis model rats was observed.To provide data and theoretical support for the clinical use of Bushen Huoxue Decoction in the treatment of glucocorticoid-induced osteoporosis.Methods1.Grouping: 30 male SD rats were adaptively fed for 7 day and then divided into 3 groups according to the random number table method,namely blank group,model group and traditional Chinese Medicine Group,with 10 rats in each group.2.Modeling:The rats in the three groups were given gluteal muscle injection,twice a week,for 8 weeks;the blank group was given an equal volume of 0.9% sodium chloride injection(2.5ml/kg);the model group and the traditional Chinese medicine group were given dexamethasone sodium phosphate Injection(2.5mg/kg).3.Administration: After the modeling,start to gavage,10:00-11:00 in the morning and16:00-17:00 in the afternoon,twice a day,the blank group and the model group were given equal volume 0.9% sodium chloride injection was given by gavage,the traditional Chinese medicine group was given Bushen Huoxue decoction 2ml/100 g by gavage,and the materials were collected after continuous gavage for 8 weeks.4.Material collection: After the gavage,the rats were anesthetized with 10% chloral hydrate,and the material was collected after the anesthesia took effect.5.Observation indicators:(1)Observe the general conditions of the rats in each group.(2)Dual energy X-ray absorptiometry was used to measure the bone mineral density(BMD)of lumbar vertebrae in rats.(3)Observe the changes of body weight and growth rate of rats in each group during the experiment;observe the wet weight of testis and kidney of rats after sampling.(4)BALP,BGP and T content were detected by ELISA.(5)Hematoxylin-eosin staining(HE)was used to observe the pathological bone structure of rats in each group.6.Statistical analysis was performed after the experimental data was obtained.Results1.General conditions: After modeling,the vital signs of the rats in each group were stable,and there was no obvious redness,swelling,oozing,or bleeding at the injection site of the gluteal muscle.During the period of ulceration,modeling and administration,none of the rats died.After 8 weeks of modeling,compared with the blank group,the rats in the model group and the traditional Chinese medicine group showed "kidney deficiency and blood stasis syndrome".After the intervention of traditional Chinese medicine,compared with the model group,the performance of "kidney deficiency and blood stasis syndrome" of the rats in the traditional Chinese medicine group was significantly improved.improve.2.BMD: After gavage,the rats in each group were tested for BMD of the isolated lumbar vertebrae.Compared with the blank group,the lumbar vertebrae BMD of the rats in the model group decreased significantly(P<0.01);Compared with the model group,The lumbar vertebrae BMD value of the rats in thetraditional Chinese medicine group increased significantly(P<0.01).3.Body weight and growth rate of rats:After modeling,compared with the blank group,the body weight of the rats in the model group was significantly reduced(P<0.01),and compared with the model group,the body weight of the rats in the traditional Chinese medicine group was not statistically significant(P>0.05).after gavage,compared with the blank group,the body weight of the rats in the model group decreased significantly(P<0.05),and compared with the model group,the body weight of the rats in the traditional Chinese medicine group increased significantly(P<0.05).During the experiment,the body weight of rats in each group increased to varying degrees.Compared with the blank group,the body weight growth rate of the model group decreased(P<0.01),and the growth rate of the traditional Chinese medicine group increased compared with the model group(P<0.05).4.Testis and kidney wet weight: Compared with the blank group,the testis and kidney wet weight in the model group decreased(P<0.05);compared with the model group,the testis and kidney wet weight increased in the traditional Chinese medicine group(P<0.05).5.Serum BALP,BGP,T Value Contents: After gavage,compared with the blank group,the serum BALP,BGP and T contents of the rats in the model group were decreased significantly(P<0.01).Compared with the model group,the contents of serum BALP,BGP and T in the traditional Chinese medicine group increased(P<0.05).6.Morphology of lumbar vertebrae in rats under HE staining: Compared with the rats in the blank group,the number and quality of trabecular bone in the model group were significantly decreased,and the osteoblasts on the trabecular bone were significantly decreased;Compared with model group,the number and quality of trabecular bone in the traditional Chinese medicine group were significantly improved,and the osteoblasts on the trabecular bone were significantly increased in traditional Chinese medicine group.Conclusion1.Applying the modeling method of "Dexamethasone Sodium Phosphate Injection2.5mg/kg,2 times/week for 8 consecutive weeks" in the gluteal muscle,the rat model of GC-induced osteoporosis can be successfully established,and it is safe and Efficient.2.Bushen Huoxue Decoction can increase the bone mineral density of GC-induced osteoporosis rats.3.Bushen Huoxue Decoction can improve bone metabolism in GC-induced osteoporosis rats,promote bone formation,increase the number of lumbar trabecular bone,and improve bone trabecular quality,so as to achieve the purpose of anti-GIOP. |
