| Purpose : The medical cases of ancient doctors who treated the syndrome of cold-dampness trapping the spleen were summarized,sorted out and collected,and the dialectics and prescription rules of cold-dampness trapping the spleen were analyzed through data mining,so as to provide reference for the modern clinical differentiation of cold-dampness trapping the spleen.Through the network pharmacology method,the core active substances,targets and specific mechanisms of the core prescription Houpu Wenzhong decoction in the treatment of irritable bowel syndrome were discussed,so as to lay a theoretical foundation and direction for the experimental verification of the treatment of irritable bowel syndrome in the future.Material and method:This paper contains two parts.In the first part,the distribution of medical cases in ancient books for the treatment of cold-dampness trapping spleen syndrome was analyzed and drawn into a table through data mining.The composition,nature,flavor,and meridian of Chinese herbs were summarized and a database was established to analyze their frequency and frequency.Apriori algorithm in SPSS Modeler 18.0 software was used to analyze the association rules of the obtained data,and the results were sorted in descending order according to the support degree.The relationship between drugs could be observed more intuitively through the network diagram drawn.Through the comprehensive analysis of association rule algorithm and cluster analysis,the core prescription of Houpu Wenzhong Decoction was obtained and selected.In the second part,data mining technology was used to query the prescription active ingredients and targets of Honpu Wenzhong decoction through the TCMSP database,and to query the targets of irritable bowel syndrome and remove the weight through the disease gene target database Gene Cards,OMIM and TTD.The intersection of the prescription targets of Houpu Wenzhong Decoction and the IBS targets is a potential target for prescription treatment of IBS.Cytoscape software was used to construct the network diagram of "Houpu Wentchong decoction-irritable bowel syndrome-targets",and the core targets were determined by PPI network diagram.Metascape software was used to perform GO enrichment analysis and KEGG enrichment analysis of therapeutic targets,and molecular docking technology was used to docking active ingredients with protein targets.It can be intuitively judged whether the compound can stably bind to the target protein in human body.Results:1 Data mining resultsA total of 485 prescriptions were obtained from 130 ancient books,and most of them were distributed in the Song,Ming and Qing Dynasties.There were 174 Chinese herbs in 485 prescriptions in the database,of which 12 were found with drug frequency ≥20% : Licorice,Tangerine peel,Atractylodes atractylodes,dried ginger,ginseng,Poria,magnolia officinalis,Aconite,cinnamon,Pinellia,woody and cloac.The herbs mainly belong to the spleen,stomach and lung meridians,among which the herbs have the most pungent,bitter and sweet flavors.The medicinal properties are mainly warm,followed by cold and smooth ones.According to the Chinese Materia Medica,they could be classified into 21 categories,among which there were 27 herbs for tonifying deficiency,with a frequency of 839,followed by 13 herbs for warming,with a frequency of 724.A total of 26 strong association rules were obtained by Apriori algorithm.According to the descending order of confidence,the highest confidence was ginseng,Atractylodes rhizoma → Licorice,followed by Magnolia officinalis,licorice → Tangerine peel,etc.The correlation results showed that the core drugs were: Atractylodes rhizoma atractylodes rhizoma,licorice,woody incense,dried ginger,Magnolia officinalis,Pinellia pinellia,ginseng,tangerine peel,aconite,Poria,cinnamon.Based on the analysis of the results,the basic prescriptions were Houpu Wenzhong decoction,Sijunzi decoction,Erchen decoction,etc.When the frequency of drugs was more than 10 times,the cluster analysis was performed,and the core prescriptions were obtained by combining the results of drug efficacy and clinical characteristics.The first prescription was zedoary turmeric,Sanleng,green peel,ferula,piper and pepper.Prescription 2: Areca nut,Fructus aurantii,Ligusticum chuanxiong,fennel,Fructus vulgaris,galangal,Chebula,almonds,cardamom,rhizome kernels,cubeb,nutmeg,clove,woody,grass cardamom,evodia;Recipe 3:Wuzhuyu,Asarum,cinnamon,Grandiflorum,malt,Shenqu,bitter orange;Recipe 4:rhubarb,Rhizoma coptidis,aconite,dried ginger,red stone fat,Sichuan pepper,Angelica;Prescription 5: jujube,ginger,cinnamomum sprigs,paeoniflora;Prescription 6: white lentils,yam,papaya,Atractylodes,ginseng,licorice,Poria,astragalus;Prescription seven: Rhizoma alismatis,Zhuling,Atractylodes atractylodes,Tangerine peel,Magnoliae officinalis,grass fruit,Pinellia pinellia,Agastache rugosa,large abdominal peel.2 Network pharmacology resultsA total of 137 active ingredients,2307 potential targets,4172 targets related to IBS,199 targets of prescription and disease were obtained,and 5 compounds with more than 30 targets were identified,including quercetin,enteringin,luteolin,kaempferol and 7-methoxy-2-methyl isoflavone.The top five gene targets with the highest correlation value were NFKB1 A,HMOX1,RELA,MAPK1,and BCL2L1.Enrichment results There were 2410 GO entries,and 209 signaling pathways related to the treatment of irritable bowel syndrome were obtained by KEGG pathway enrichment analysis.The core targets and main active components were found to have strong connectivity by molecular docking technology.Conclusion:Through the data mining of ancient books,it is found that in the treatment process of cold-dampness trapping spleen syndrome,ancient doctors paid more attention to the regulation of spleen and stomach,taking into account the lung and kidney.In terms of the nature and taste of drugs,they mostly used spicy,bitter and sweet drugs to warm the middle to dissipate cold dryness and dampness.In the treatment of cold-dampness trapping spleen syndrome,they commonly used drugs such as tonifying deficiency,warming,regulating qi and enhancing hydration and dampness.Based on the analysis of association rules of drugs used by ancient doctors for the treatment of cold-dampness trapping spleen syndrome,they often added or reduced Sijunzi decoction,Houpu Wenzhong decoction,Erchen decoction and dried ginger Fuzi decoction,etc.Through cluster analysis,a total of seven prescriptions were obtained,aiming to provide ideas and basis for the clinical treatment of cold-dampness trapping spleen syndrome.Through the network pharmacology study of Honpu Wenzhong decoction in the treatment of irritable bowel syndrome,a number of potential core targets for the treatment of irritable bowel syndrome have been screened,which can affect oxidative stress,cellular inflammatory response,cell proliferation,differentiation and other biological processes,and the core targets and pathways have been verified,which lays a certain foundation for clinical application,but this conclusion needs further experimental verification. |
