| Objective:To observe the effects of Penyan pills on rats in a pelvic inflammatory disease sequelae(SPID)model through an experimental study,with an eye on ferroptosis and fibrosis perspective,and to provide a new target for the treatment of SPID,and to provide theoretical and experimental support for the prevention and treatment of SPID by traditional Chinese medicine.Methods:Forty SPF-rated SD rats were used in this study,and after observation using the vaginal cytological smear method,membranes were created in the rat’s motility cycle.40 SPID rats were divided into 10 in the sham-operated group and 30 in the modeling group.The sham-operated group underwent sham surgery,and the modeling group was established by the chemical injury method,is also known as phenol slurry method,and the modeling group was randomly divided into three groups:modeling group,Penyan pills group and azithromycin group,10 rats/group.The rats in each group were treated with gavage for 28 days,and the general status and weight changes were observed during the treatment period.4 weeks later,specimens of uterus,fallopian tubes and ovaries were collected for examination.The morphological changes of pelvic tissues were observed by HE staining method,and the mRNA expression levels of GPX4,NF-κB,TGF-β1 and SMAD7 in uterine tissues of SPID rats were detected by RT-qPCR.Results:1.Gross morphology:compared with the sham-operated rats,the thickened and swollen right uterus and fallopian tube of the model group rats and fluid accumulation were obvious,with altered texture and reduced elasticity,visible adhesions with surrounding tissues and pelvic fluid accumulation,which proved the success of membrane-making;the uterine fallopian tube of the Penyan pills group and the azithromycin group rats were well-proportioned in appearance,without edema and textural changes,with light red color and elasticity.2.HE staining:the uterus of the sham-operated group rats The tissue structure was clear,neatly arranged,without edema,without necrosis and detachment,with mild hemorrhage,a very small amount of inflammatory cell infiltration and a slight increase in the number of fibroblasts in the tissue.In the model group,the structure of the uterine cavity wall was abnormal,the arrangement was changed,a large number of endometrial glands were seen,the glands were enlarged and irregular,a small amount of hemorrhage was seen in the tissue vessels,a slight increase in the number of fibroblasts was seen,and a small number of inflammatory cells were infiltrated.This suggests that the membrane was successfully formed in rats using the phenol plasma chemical injury method.In the azithromycin group,the uterine tissue structure and morphology were basically restored to normal,neatly arranged,no degeneration and detachment,no obvious edema,a small amount of hemorrhage,a small increase in the number of fibroblasts,and the muscle layer and plasma membrane were basically intact.In the Penyan pills group,the uterine tissue structure and morphology were basically normalized,neatly arranged,no degeneration and detachment,slight increase in the number of fibroblasts,no obvious edema,and a small amount of hemorrhage could be seen.3.Compared with the model group,the expression of TGF-β1 and NF-κB was decreased in the Penyan pills group and azithromycin group,while the expression of GPX4 and Smad7 was increased(P<0.05)Conclusion:1.The phenol plasma chemical injury method can successfully replicate the SPID rat model.2.The formation of SPID may be related to tissue fibrosis and iron death.3.Penyan pills can repair the pelvic tissue morphology and improve pelvic adhesions in SPID rats.4.Penyan pills can improve iron death,promote iron death,and promote the proliferation and growth of SPID rats by down-regulating the expression of TGF-β1 and NF-κB and up-regulating the expression of GPX4 and Smad7.The mechanism of action of pelvic inflammatory pills and azithromycin in regulating iron death in SPID rats may be similar. |
