Human monoclonal antibodies (Hmab) do have some advantages over the counterparts of murine's as we know. But in contrast, unfortunately, the preparation of Hmab is much more difficult. Scientists have never stopped struggling on solving certain problems since the emerging of Hmab, such as the shortage of primed human Blymphocytes, low fusion frequency, poor cloning efficiency and limited antibody production. A new method for producing Hmab is the genetic engineering antibody technique which is presumed to be promising and productive. But the fact is that this method has not yet been sophisticated upto now and can not be used as a routine procedure in a period of time. The major approaches for raising Hmab today that we have been, using for more than a decade are still EBV transformation, hybridoma and EBV-hybridoma techniques even they are not as good as expected.Int erleukin-11(IL-11) is a novel cytokine which has been found effective on promoting the antigen-specific response of human Blymphocytes. In this thesis, we constructed a retroviral vector PDoR—IL11 and succeeded to transfer...
