To break through difficulties studying in aqueous enzymatic extraction oil, the new methods and related mechanisms were studied with soybean containing low oil as material to complete the simultaneous utilization oil and protein of oilseed. First , the technology of aqueous extraction oil and protein was studied by condition test and orthogonal test. The optimum conditions of extraction oil and protein from soybean by water is crushing the material to 20 mesh of particle size , the ratio of solid to liquid 1:4, soaking in water of pH9.0 and 500C for 1 .5h, grinding with water to less then 100 mesh of particle size, filtering and washing ,the 96%total oils and 95% total protein could be extracted into soybean milk. Hydrolyzing the soybean milk by 1398 neutral protease under different conditions, the oil has different distribution between in milk and solid by centrifugal separation, according to which, better method of enzymatic extraction oil can be designed. To decline the amount of enzyme and to improve the efficiency of extraction oil, organic solvent was used in extracting oil from soybean milk, Three methods for aqueous enzymatic hydrolysis were studied, Soybean milk directly hydrolyzed by enzyme, enzymatic hydrolysis in protein milk isolated from soybean milk or soybean milk pre-hydrolyzed and protein isolated to enzymatic hydrolysis. Amount of protease used in three methods was 8401U, 3601U, and 5 1OIU per gram soybean, respectively. After enzymatic hydrolysis the extracting oil yield was 80.2%, 70.1% and 84.5%. The highest centrifugal force demanded was 1400g. Compared to papers, three aspects including amount of enzyme, centrifugal force and yield of oil extraction have been broken through. Conditions of oil extraction and emulsification controlling during extraction oil from soybean milk were studied systematically . The experiment results showed that both could be unified well. According to the extraction fraction the three methods were compared. The above research can provide guide for application. It is the first time, factors of affecting reaction velocity of 1398 neutral protease in soybean protein milk system were investigated. The results showed that the particle size and concentration of the solid in milk influenced on reaction velocity obviously. There is great error between experiment and Michaelis- Menten equation for enzymatic reaction velocity in milk system.
