Much attention has been focused on natural polysaccharides because of its important status in life sciences. Fructus lycii, the fruit of Lycium barbarum L, is a well-known traditional Chinese medicine and food, which has multiple pharmacological activities and bioactivities, Lycium barbarum polysaccharides are the main function ingredients of fructus lycii. Because traditional Chinese herb has not been studied by modern science and technology until ten years ago, it drops behind now. To develop traditional Chinese herb, forward position technology on biology must be applied. In this study, the characteristic, structure and bioactivities of LBP were studied. And to study LBP's function mechanism, laser scanning confocal microscope, flow cytometer and gene express were preformed in this paper. The main results are as follows:1 The characteristic and structure of LBPFructus lycii was chopped into pieces and extracted by solvent I for 4 times. Then the residue was extracted by solvent II for 4 times. The solution was concentrated by vacuum concentrator and was treated by polyamide column. After have been vacuum concentrated, freeze-dried, extracted by solution IE for 4 times and vacuum dried, LBP was gained. The yield of total LBP was 2.64% 0.12%. And the purity of LBP is 94.3% 1.5%Polyamide column could separate protein from LBP. After have been treated by polyamide column for 1 time, the content of protein in LBP decreased from 14.7% 0.2% to 4.7% 0.1%. The regain ratio of LBP on polyamide column is 98.7% 0.5%. And the exchange capacity of polyamide column to protein is 5.2 0.1 (mg'g-1 ).LBP was separated by DEAE-Cellulose column with the eluent of distilled water, 0.05 mol-L-1, 0.10 mol-L-1 and 0.25 mol-L-1 NaCl, respectively. The last part of LBP separated by DEAE-Cellulose was purified by sephadex G-200 column with distilled water aseluent and the LBP-4a was gained. The monosaccharides in LBP-4 identified by GC were xylose (Xyl), galactose (Gal), mannose (Man), Rhamnose (Rha), glucose (Glu) and arabinose (Ara) with the molar ratio of 4.45: 1.20: 1.0: 0.98: 0.90: 0.85. The content of calcium, magnesium, zinc and iron in LBP-4a were 127.9070 mg-g"1, 32.3953 mg-g"1, 0.3845 mg-g"1 and 0.2930 mg-g"1, respectively. The content of galacturnic acid in LBP-4a was 55.45% while the content of protein is 3.80%.According to IR spectra, it contains -COOH group, -NH- group, -OH group, -NH2 group and P -D-glucose. The backbone of sugar residues chain in LBP-4a were contained l-6 indican bond according to periodate oxidation and the results of -elimination reaction indicated that the chain of polysaccharides and protein were connected by O-linked chemical bond. The photograph of LBP-4a got by laser scanning confocal microscope and scanning electron microscope showed that the LBP-4a combined both with each other and with itself in its crystalloid. The crystalloid was flake and anisotropic.2 Effects of LBP on tumour cellsLBP-4 has the effects of antitumour and could induce apoptosis. LBP-4 at the dose of 50 mg-L-1,100 mg-L-1,200 mg-L-1 and 400 mg-L-1 could inhibit the growth of QGY-7703 cells and could enhance apoptosis index. The morphological observation showed that the karyon of QGY-7703 cells shrinked and the surface of karyon became coarse.The QGY-7703 cells were blocked in S-phase and the concentration of calcium in cytoplasm was enhanced according to the results of flow cytometer assay and determination of the calcium concentration by laser scanning confocal microscope.3 Effects of LBP on growthLBP-4 could be valuable to growth. After taking LBP-4 for 3 weeks, the increase of body weight slowed down and the food conversion rate enhanced. While the thymus index enhanced, the liver index and spleen index reduced.LBP-4 is benefit to absorbing of metal elements. And it could help to reduce the resistance of iron to zinc and enhance both the zinc and iron in crural bone and could enhance calcium and zinc in pygal muscle of mice.4 Effects of LBP on obesityLBP could cure obese mice...
