Rhodiola L , as an adaptability plant living in specific circumstance, has many biologic activities. It's also one of rare plant sources, which can be edible and be as medicine. There are 90 more varieties all over the world, while over 70 in China. Up to date, only about 20 varieties have been studied on their chemical ingredients and/or some biologic activities. Many researches have proved that most of Rhodiola L. contain the effective ingredient-salidroside. The effective ingredients of the same variety of Rhodiola L. planting in different territory are different.Research about the effective ingredients of Rhodiola henryi S.H.Fu (RH) have been just carried out. And very few studies on the biologic activities of RH have been found reported.The subjects, Rhodiola henryi was originated from forestry centre of Shennongjia, Hubei, P. R. China. This paper aimed at systematic studies of effective ingredients, polysaccharides from R.henryi and its some biologic activities.Furthermore,we studied and developed the functional food using these effective ingredients.1. The effects on the extract rate of the effective ingredients in different factors were investigated, such as extraction temperature ,time, ethanol content, PH and so on,and the optimal condition of extracting process was gained:70% ethanol/ RH : 16:1(w/w); Extraction temperature: 80℃;Re-extraction times: 3 times; Extraction time: 60 min. every time.Using these parameters above,the content of effective component (salidroside) was 1.1% in the dry extract.After the ethanol extract was isolated by 70% ethanol at 80℃ extracting and volatiliced, the extracts were successively got by petroleum ether, chloroform,ethyl acetate and n-butyl alcohol extracting and volatilized. The n-butyl alcohol extract was purified by Al2O3 and silica gel column chromatography using chloroform and ethanol(6:4; 4:6), then the obtained solution and crystallized by CH3CHCl2 and ethanol, the white acicular crystal was obtained and identified as salidroside by modern analytical technical, such as thin layer chromatography(TLC), UV, IR ,NMR, which were first isolated from R. henryi in the world.And the other two ingredients were obtained by polyamide resin from the ethyl acetate extract. One was being identified. The other ingredients, a light yellow crystal, was identified as Tyrosol ,the precursor of Salidroside ,it is also an effective ingredient.2. Biologic activities studies on the ethanol extract from R. henryi (RHE) had been carried out. The result suggest: effects of RHE (300mg/kg.bw) on normal mice and mice of memory impediment model were obvious and had significantly differences compared with control groups. Effect of RHE (200-400mg/kg.bw) on the survival time in mice exposed to hypoxia were also obvious differences with control groups,and were statistically significant (P <0.05).3. The R. henryi polysaccharides (RHP) were isolated from the dry roote-stem of R. henryi after ethanol extracting, by water-soaking the residue and extracting. RHP-Ⅰ, RHP-Ⅱ and RHP-Ⅲ were obtained indeperelently by 30%, 50%, 70% ethanol precipitating . Using RHP-Ⅲ, eliminating albumen with the method of Sevag, decolorating. dialysis and freeze drying, and then,it was purified by DEAE-cellulose column chromatography(by 0.1M Nacl,0.5M Nacl). As purified polysaccharide-RHP-ⅢS was obtained and identified by Sephadex G-200 column chromatography.The typical absorption of RHP/RHP-ⅢS were shown in UV and IR spectrum. The result analysed by paper chromatography shows these compositions of monosaccharides are arahinose(Ara), rhamnose (Rha), glucose (Glc) and a monosaccharide which will be identified.4. Study on the biologic activity of the R. henryi polysaccharides (RHP) Experiments about anti-fatigue effect of RHP showed: RHP can obviously prolongswimming time in the burdened mice, and it can also decrease the level of BUN and BLA in mice.
