Derivatization with fluorescent molecules is an important way of increasing the detection sensitivity of chromatography. This dissertatioin is focused on the development of novel fluorescent reagents and labeling methods. Several fluorescent reagents have been synthesized and the relevant HPLC methods have been established.Quantum chemistry study on the optimized geometries and fluorescence excitation spectra of carbazoles and acridones has been carried out. With the logical design of reaction way, two novel fluorescent reagents for carbonyl compounds have been firstly synthesized. They are 2-(9-carbazole) -ethoxycarbonyl hydrazide (CEOC-Hydrazide), which is based on the fluorophore of carbazole and 1,3,5,7-tetramethyl-8-phenyl-(4-(3-epoxypropoxy))– difluoroboradiaza-s-indacence (TMEPBODIPY), which is based on the fluorophore of boron dipyromethene. Two novel fluorescent reagents, 1,2-Benzo-3,4-dihydrocarbazole-9-ethyl p-toluoylsulphonate (BDETS) and 1,2-benzo-3,4-dihydrocarbazole-9-ethoxy-carbonylhydrazine(BCEOC-Hydrazide ), which are based on the fluorophore of 1,2-Benzo-3,4-dihydroca-rbazole have also been developed. The experimental results demonstrate that the derivatization reaction is not only easy and fast, but also helpful to improve the separation resolution and the detection sensitivity.The derivatization and HPLC methods for saccharides and formaldehyde have been studied by using CEOC-hydrazide and BCEOC-Hydrazide as fluorescent...
