In order to increase saponin physiological activity, the enzyme hydrolysis of saponin-sugarside such as glycyrrhizin and ginsenoside by enzyme, the saponin enzyme production from microbial strain, purification and characterization of the saponin enzyme were studied. And the behaviors of soybean saponin in the traditional soybean food fermentation were also studied.The glycyrrhizin was hydrolyzed by 2 N HC(?)-methanol to obtain aglycon, and the aglycon yield was 92 %. In the isolation of glycyrrhizin by the resin, the absorbent amount was 209.9 mg/g for D101 resin, and 256.0 mg/g for HP-20 resin, and the glycyrrhizin could be eluted by the 50 % alcohol. In the purification, the glycyrrhizin yield was 60 % for the D101 resin, and 86 % for HP-20 resin. The Aspegillus sp.48 strain from 8 type strains produced highly the saponin enzyme which can hydrolyze the glycyrrhizin turn to monoglucuronide-glycyrrhizin ( Its sweetness was 1000 times of the sugar), and sp.39 strain produced the enzyme which hydrolyze the glycyrrhizin to the aglycon. The optimal enzyme production for the two strains was both obtained after fermented for 40 hours. The supernatant culture was precipitated by the (NH4)2SO4 powder, dialyzed, and purified by a column of DEAE-Cellulose DE52 to gain pure enzyme. In the purification, the yield was 3.1 % for the sp.48 strain, and 7.2 % for sp.39 strain. The enzyme molecular weight was 60,000 for the sp.48 strain, and 42,000 for the sp.39 strain in the SDS polyacrylamide gel electrophoresis. The purified two enzymes can not hydrolyze the p-nitrophenyl-β-glucuronide and p-phenolphthalein-β-glucuronide, and the enzyme from sp.48 strain hydrolyzed glycyrrhizin to the monoglucuronide-saponin which sweetness was 1000 times of the sugar, and the alycon was slightly produced aglycon in the long reaction. And the enzyme from the sp.39 strain hydholyzed the glycyrrhizin to the aglycon. When the glycyrrhizin was hydrolyzed with culture of sp.48 strain, the yield of monoglucuronide-saponin having higher sweetness was 10 %, the non-reacted glycyrrhizin could be retrieved. And the production can be isolated by the method of resin column from the culture.The Aspergillus sp.48g and sp.848g strain of the 8 type strains highly produced...
