Nano lipid carrier refers to various organized molecular assemblies which assembled by lipids.It develops very fast in recent years and is applied mainly in drug carrier and sensor carrier.Objestive:To prepare kinds of nano lipid carrier,research the formulas,preparation technology and characters of common,magnetic and photopolymerization liposomes and lipid LB film,and to take the exploratory development of lipid drug carrier and sensor carrier.Methods:For lipid drug carrier,the enbedding ratio as an index was evaluated to the optimizing preparation technology with an orthogonal design.Common liposome and magnetic liposome were prepared with reverse phase evaporation.For sensor carrier,the specific recognition was used as an index to evaluate and to prepare photopolymerization liposome with sonication and to prepare lipid LB film with level contact method. Their shapes,sizes,Zeta electric potential,magnetic propertied,crystal analysis etc.were detected with atomic force microscope,transmission electron microscope,nanosizer,X-ray diffractometer,vibrate sample magnetometer,HPLC.Results:Materials in preparation,tissues and serum do not trouble the detection of selected medicine in HPLC at selected wavelength.The optimal preparation technology of Clindamycin Phosphate liposome as follow:taking ultrasound for 6 min,evaporation temperature was 35℃, drug:phospholipids as 3:5,phospholipids:cholesterol as 5:1, phospholipids:mycose as 2:1.To determine three batch number liposome, the enbedding ratio were 52.26%,50.13%and 53.75%.The size of magnetic nano-particle by improved co- precipitation was varied from 10 to 50nm.The temperature increase value was positively correlated with its content and different current.Semustine magnetic liposome is with Zeta electric potential as-37.1±0.60mv and pH value as 7.41 with superparamagnetism and strong magnetism,embedding ratio as 73.72%. AUC value of 24 hours at rectum in Semustine magnetic liposome group was 20.9 times of that of free drug group,and 3.6 times of that of common Semustine liposome group.Its concentrations in kidney were lower than those in free drug group.Its rectum-targeted ratio was 59.3%. The optimal preparation technology of photopolymerization liposome as follow:lipids concentration was 1mM,taking ultrasound for 12 min, illumination distance was 1cm and the time was 60min.The color of photopolymerization liposome solution was dark blue,the maximum absorption peak of visible absorption band was about 640nm,mean size was 100-200nm.The prepared colorimetric immunosensor could detect schistosome antibody quickly.The color of the liposome solution turned from dark blue to prunosus in 2min,and the maximum absorption peak of visible absorption band transmited from 640nm to 540nm,color response value was 32%.Lipid LB film was prepared with level contact method. The particle diameter of the gold colloid which was prepared by trisodium citrate reduction was about 20nm.The prepared piezoelectric immunosensor could detect the antibody in the blood sample.Conclusions:Lipid drug carrier had a simple preparation condition and good stability.It could decrease the nephrotoxicity of the drug and increase drug availability.The preparation method of the sensor carrier was special and simple;the specificity of immunosensor was powerful and it implemented the rapid detection of schistosome antibody.These prepared nano lipid carriers were reached objective in this research.
