Chinese ring-necked pheasant has the special fleshy flavor,which has more quantity now.So it will become the emphases in Chinese ring-necked pheasant research how to keep these special characteristics such as the fleshy flavor and how to further improve production performance and fleshy flavor.The content of intramuscular fat(IMF) and inosinemonophosphateacid(IMP) are closed related with muscle tenderness and flavor,for which IMF and IMP are the main merits that can affect meat quality and have some major function which affect the content of IMF and IMP content.We consider these genes as the potential candidate genes of fleshy flavor.In this experiment,we choose A-FABP,H-FABP,LPL,ADSL and ATIC genes of the Chinese ring-necked pheasants as candidate genes of meat quality,then we did analysis by using PCR-SSCP,Real-time PCR and bioinformatics,The following conclusions were drawn through these studies:1.The IMF content of the Chinese ring-necked pheasant in breast and thigh muscle were lower than those of Jilin Sanhuang chicken and AA chicken.2.The nucleotide and amino acid sequences of Chinese ring-necked pheasant A-FABP,H-FABP,LPL,ADSL and ATIC have the closest phylogenetic relationships with the poultry.A-FABP and H-FABP genes have Lipocalin family domain.LPL gene has Esterase-lipase family domain,ADSL gene has Lyase family domain,ATIC gene has AICARFT family domain.3.Four mutations were detected in A-FABP gene,which have a significant effect on live weight,carcass weight and IMF content in breast muscle(P<0.05),Four mutations were detected in H-FABP gene,which have a significant effect on IMF content in breast muscle(P<0.05),Three mutations were detected in LPL gene,which have a significant effect on live weight and carcass weight(P<0.05),A mutation was detected in ADSL gene,which has a significant effect on carcass traits(P<0.05),A mutation was detected in ATIC gene,which has a significant effect on the IMF content in breast muscle(P<0.05).If we merge two positions of the Chinese ring-necked pheasant populations,AAAA genotype has the highest IMF content in breast muscle.4.A-FABP gene has 16 haplotypes,in which haplotype combination of H9H11 and H1H4 can improve carcass traits and IMF content,H-FABP gene has 14 haplotypes,in which the haplotype combination of H3H6 and H4H6 can improve carcass traits and IMF content,LPL gene has 7 haplotypes,in which haplotype combination of H1H5,H3H6,H1H1 and H1H2 can improve carcass traits.5. A-FABP,LPL,ADSL and ATIC genes have the highest expression when they grow about 150d. H-FABP has the highest expression when they grow about 60d.A-FABP gene can express highly in the adipose tissue,H-FABP gene can express highly in the heart,LPL and ADSL genes can express highly in the muscle tissue,the ATIC gene can express highly in the liver.6. Significant negative correlation was deteceed between A-FABP gene expression in the Liver and IMF content in breast muscle,significant positive correlation was deteceed between H-FABP gene expression in the liver and IMF content in thigh muscle,Significant negative correlation was deteceed between H-FABP gene expression in the adipose tissue and IMF content in thigh muscle,significant negative correlation was deteceed between LPL gene expression in the adipose tissue and IMF content in thigh muscle,significant negative correlation was deteceed between ADSL gene expression and live weight.
