Study on the Immunology and Apoptosis of Chicken and Tumorigenic Mouse Induced By SAg-SEB and MDVABSTRACTIn recent years, it is known that superantigens (SAgs) are molecules produced by bacteria and viruses. SAgs are characterized by their abitity to polyclonally activate T cells through the Vp element of the T-cell receptor (TCR). The consequences of SAg exposure range from the induction of T cell proliferation and cytokine release to immuno suppression and tolerance.It is now identified that SAgs induce cell mediated immune response, anti-tumore reaction and adjuvant function. But the machinism is not reported in birds and it is not known whether simillar machinism is in birds. The immune machinism of chicken induced by SAg SEE will be first discussed in this study. The immune machinism of MD in chickens and its relativity with SAg will be also measured and analysed by using Thj cytokines and apoptosis mearure at first time; The anti-tumor action of SAg-SEB and MDV in Tumorigenic Balb/c mouse, the future using of SAg in MD control and the SAg funtion of MDV is also discussed. These results showed as follow:1. SAg-SEB increased the release of IL-2, IFN-Y and TNF of thymocyte, spleno-cyte and bone marrow macrophage in vitro; and also increased the cell mediated immune response and anti-tumor ability, induce apoptosis, strengthen anti-disease power of chichen including that higher dose of SEB induced a transient increase and followed decrease of cytokine release, lower dose of SEB induced a slow increase and decrease of cytokine release.2. The inoculation of MDV in general indued to decrease the production of IL-2, IFN-7桾hi cytokines in vitro, and there are differences between spleen and Thymus lymphocyte on the production of IL-2 in vitro, only thymus lymphocyte IL ? prodution increased transient in early time. The cytotoxity TNF induced in thymus and spleen lymphocyte and bone marrow macrophage shown a simillar curve of "V" shape. MDV infection cause cell 梞ediated immune declines.3. The death ratio of MDV infected chicken is depressed following exposure to SEB at 10 days PI and shown that SEB can raise the ability of anti-infection and anti-disease.4. Both SEB and MDV induce the apoptosis of thymus and spleen lymphocytes in chicken. The apoptosis of speen lymphocyte is more remarkable than thymus following exposure to SEB. and it is contrary following exposure to MDV. These results shown that SEB mainly induce mature T cell to apoptosis and MDV mainly induce immature T cell to apoptosis.5- The apoptic change in tissue distribution of thymus and spleen of chicken is simil-lar after exposure to SEB and MDV, mainly appeared in T cell and some macrophage; The morphic apoptosis changes is in different stages, including condensed nuclei and apoptic body formation, the apoptic cell shown two disorganization forms?in-macrophage and out-macrophage, the former is main form, all of these indicated that there are other machinisms in apoptic cell clearing except for macrophage. In splenocyte apoptosis process in the metabolism and protein synthesis was strengthened in transient time by SEB; Thymocyte necrosis often appeared in the late stage of MDV inoculation.6. The ratio of tumor/body weight decreased following Balb/c mouse exposure to SEB or MDV, SEB is more markable, which indicated that SAG-SEB has more powerful effect on anti-tumor ability; the ratio of spleen/body weight is increased by MDV but it is not by SEB. MDV maily cause splenocyte apoptosis, but tumor cell apoptosis is low-grade; Both tumor cell and splenocyte shown remarbable apoptosis following exposure to SEB. At the same time, cell necrosis, proliferation and transformation also be induced by SEB and MDV with apoptosis.
