Viral disease is a natural and common disease,which is a disease endangers the health of livestock and poultry.The tumor disease causes the livestock and poultry production performance to drop,even death,causes the economic loss to the farmer.T cell lymphoma in poultry is mainly caused by Marek's disease,the diagnosis method is mainly based on the extraction of virus samples in poultry,to judge the virus.With the continuous variation of the virus and the development of Marek's disease will be more and more toxic,the role of vaccination is very limited,the prevention and control of Marek's disease will face more challenges in the future.Chinese medicine has unique advantages in anti-virus,multi component,multi way,multi target,not easy to produce drug resistance.According to pharmacological research reports,baicalin has antimicrobial,antiviral,anti allergy,sedative,gallbladder and liver function,has good clinical application value.In order to study the biological activity of baicalin,this study intends to explore the extraction process of baicalin,to establish a quality standard of baicalin;in Marek's disease model of Baicalin in vitro and in vivo against Marek's disease virus test system of baicalin antiviral activity,and to further elucidate its mechanism,provide the basis for the flavonoids obtained can effectively control the virus induced tumor disease for clinical and experimental results are as follows:1.The method for the determination of baicalin content was established by UV spectrophotometry,the content of baicalin was determined by 278 nm,and the standard curve of concentration and absorbance was established.The standard curve of baicalin was A=0.0625C+0.005,R2=0.9995.Complex requirements of precision,stability,repeatability and recovery.2.The extraction process by orthogonal design of baicalin by hot water extraction,by optimizing the extraction factors,orthogonal test of three factors and three levels,the variance analysis results showed that the extraction times is 2,solid-liquid ratio was 1:25,extraction time was 50 min,the highest rate of baicalin,reached 20.36%.3.Baicalin in chick embryo fibroblast?CEF?determination of the maximum safe concentration of baicalin could promote CEF cell adhesion and growth of CEF cells and the role of timeliness in medium and low concentration,application of the CPE method and MTT method,the measured maximum safe concentration of baicalin was 312.5 CEF g/mL.4.MDV cell half infection(TCID50)determination: MDV of half the infection amount of TCID50 was 10-4.5/0.1 mL.5.The anti MDV effect of Baicalin in vitro proliferation,infection blocking and killing three different routes of administration,concentrations of baicalin has better antiviral effect in the 78.13312.5 range of g/mL,and the infection is under way to block the baicalin concentration of 156.25312.5 g/mL can promote cell growth,direct inactivation under the way of baicalin concentration of 312.5 g/mL could promote the growth of cells.6.The anti MDV effect of Baicalin in vivo results:?1?through the statistics of baicalin is high,medium and low dose group,ribavirin group,virus control group,normal control group,the number of days the death of tumor incidence and changes of chicken weight before and after the test,the obvious baicalin high dose group delay of virus the better than the positive control group and baicalin low dose group;?2?by observing the organs of each section,can also be that baicalin group can reduce the severity of MDV infection.In summary,the orthogonal method to optimize the extraction process of baicalin method is reasonable,can get high extraction rate of baicalin,clinically used for delaying Marek's virus infection,and the tumor disease as the research animal model of Marek's disease research and treatment of Baicalin on animal tumor disease is promising.
