From the developmental genetics angle, the protein comparisons from different developing stages of different organs of T-cytoplasmic male sterility (CMS) and fertility restoration in common wheat are carried on by means of IEF-SDS PAGE with an improved silver stain technique. And the results are as follows:(1) Four experimental lines have been created by intercrossing among the nearly isogenic lines of the fertility restorer,Rf3. And their genetic backgrounds are very similar except the locus of the fertility restorer, which avoids the other genes interrupting.The protein differentiation of different developing stages (meiosis and post-meiosis of microspores) of different organs (immature ears and flag leaf issues) among the four experimental lines is analyzed and the proteins related to male sterility and fertility restorer are determined preliminarily.(2) The differentiation of the protein expression between the immature ears of experimental lines has been found. The proteins related to the male sterility at the meiosis stage in the immature ears are isolated, i.e,112.621KD, 104.588KD, 98305KD(a), 98.305kD(b), 88.245kD, 60.573kD, 59.561kD, 54.132KD, 44.727kD, 36.400kD, 28.975kD, 25.506kD; and those related to retaining fertility are 45.140kD, 38.226kD, 35.851kD, 36.630kD, 33.095kD, 30.364kD; The proteins related to the male sterility at the post-meiosis of microspores are also isolated, i.e. 95.215kD, 83.396kD, and that related to retaining fertility is 42.402kD and 39.180kD. According to the results, the most active stage of genes correlated with male sterile or its fertility restoration should be at meiosis stage.(3) The four experimental lines have the same patterns of the proteins two -dimensional electrophoresis at the meiosis and post-meiosis of microspores stages, which indicates the expression of the male sterility and its fertility restoration genes has organ-special characters.(4) The two-dimensional electrophoresis fitting to the common wheat protein has been set up through many repeats. And the most important work is that we improve the IEF-SDS PAGE with diamine silver stain, whose sensitivity is high and background is low.(5) Construction of natural protein marker system has been proposed and a simple protein database consisted of 7 natural marker system has been set up. The calculating-formula of protein molecular weight has been improved. 7=-6.5057x+13.099.
