Cryo-Damage Detection Of The Spermatozoa And Its Lectin-Binding Pattern In Borderdale And Dorset Ram

The changes of enzyme (superoxide dismutase(SOD), nitric oxide synthetase(NOS) , glutathione-Stransferase (GSH-ST) , catalase (CAT) , choline esterase (CHE) , akaline phosphatase (AKP) , acid phosphatase(ACP) ,y-glutamyltransferase (y-GT) , sialic acid(SA) , acety-laminoglucosidase(NAG) , glutaminesynthetase(GS)) and the trace elements (kalium(K), natrium(Na), chlorine (Cl), calcium(Ca), phosphorous (P), magnesium(Mg)) of cryopreservation spermatozoa was determined in this study. The contents of NOS and GS increased GSH-ST, AKP, SA, SOD, CAT, CHE and y -GT decreased evidently after the freezing. The contents of Na, Cl and K showed a evident decreasing and P showed a evident increase. These kinds enzyme and trace elements may be a important indicator of the cryodamage of the ram spermatozoa. Other enzyme and trace elements showed no evident changes in this study.In the study on the sperm hypo-osmotic swelling test (HOS), the type of swelling spermatozoa was divided into two types, "g" type and normal swelling type. The results showed that the "g" type spermatozoon is more suitable for the detection of the membrane integrity. At the same time, the capacition in vitro of those spermatozoa sample was carried through either and the results showed that membrane integrity didn' t affected by capacitation.The results of the DNA damage detections showed that DNA may not be damaged by capacitation and the data of the single-cell gel electrophoresis (SCGE).The pattern of the lectinin in this study showed that: (l)the binding of Con A after the capacitation is more stranger than before and Wheat germ showed the similar situation. But it is not obviouslycompared with other people' s results. Otherwise, these kinds of lectin may be are useful tools to indicate the capacitation of the ram spermatozoa. (2)the pattern of Lentil and Horse gram showed a weak binding in this study and had no changes before and after capacitaion.
