Heterosis is an important approach for crop improvement; thermo-photoperiod sensitive male sterile wheat is a valuable material for heterosis study and utilization by means of two lines system. Thermo-photoperiod sensitive male sterile wheat has many distinguished advantages compared with CMS, such as"one line two-use"(sterile line and maintainer line), abundant restoring resources, and better and convenient hybrid production and high purity sterile line reproduction. However, the mechanisms of thermo-sensitive male sterility conversion were still unclear. In this study, the pollen fertility performances and the gene expression profiles between sterile spike and fertile spike of line A3017, a YS type thermo-sensitive male sterile wheat, were analyzed and compared under controlled male sterile or fertile temperature conditions by means of mRNA differential display (cDNA-AFLP and SSH), and the main results were as follows:1. The comparison of pollen fertility between male sterile and fertile spike was conducted with two lines, 310 and 312 line of A3017, cultured in growth chamber. The results showed that under the condition of male sterile (day/night temperature at 16℃/12℃), the anther volume of 310 and 312 were smaller than their normal counterparts, and the anthers could not open, even if they can open, there was no pollens dispersed, and the ratio of sterile pollen stained with I2-KI solution were 98.7% and 99.0%, respectively, the pollen germination potential were 0.7% and 0.5%, respectively, and both self seedset ratios were 0 for both lines; under the condition of male fertile (day/night temperature at 22℃/18℃for normal tillers and 25℃/20℃for regenerated tillers), the anthers of 310 were normal, while part of anthers of 312 were normal at 22℃/18℃, but there were no difference for the anthers of those two lines at 25℃/20℃, the percentage of sterile pollen stained with I2-KI solution to total pollens observed under above conditions were about 20% for both lines. The pollen germination potential increased with the ascending temperatures, which were 60.7% and 71.5% for 310, 27.4% and 63.1% for 312, respectively.
