| In order to select the conversion materials of thermo sensitive male sterile BNS, theexperiment used200common wheat line as male parent materials to carry on cross andanalyzed F1self-fruitful rate. The F2generation of cross combination BNS/XiNong9062wasanalysed with the mixed model of major gene plus polygenes for quantitative traits. Thedifferentially expressed mRNA of these proteins’ genes, namely ATPase α submit and APRT,were examined by means of fluorescence real-time quantitative PCR (qRT-PCR) taking theanthers of BNS as materials at four key developmental stages (tetrad phase, uninucleatedperiod, binucleated period and trinucleate period). The results showed that:(1) The12combination of the F1self-fruitful rate over50%with international method,the restoring rate range from51.25%to134.72%(international method),36.25%to93.05%(domestic method). The combination of the best self-fruitful were BNS/728, BNS/928,BNS/XiNong1918and BNS/11ZC05, and the self-fruitful rate of international method were134.72%,127.63%,90.84%and92.72%, respectively.(2) The fertility of the BNS was controlled by two additive-dominance-epitasis-majorgenes. Two major gene were additive effects and dominant interaction effects. Majorgeneheritability was97.08%.(3) The relative copy numbers of the ATPase α submit was sustained decline from tetradphase to binucleated period in sterile conditions, and was significant decrease as comparedwith the fertile conditions in the uninucleated period. Therefore, ATPase α submit ispositively related to the conversion of BNS sterile. And this relationship is more obvious inuninucleated period specially.(4) The relative copy numbers of APRT1gene in the sterile BNS plants was lower thanthat of fertile BNS plants from tetrad phase to trinucleate period. While the transcription levelof APRT2gene in the BNS plants kept low on the whole. The transcription level of APRTrelated genes was significant increase in trinucleate period, and the fertile BNS plants wasmore obvious than the sterile BNS plants. Therefore, the transcription level of APRT gene isprobably related to the fertility alternation in BNS wheat in trinucleate period. |
