| The effects on separation and identification of Allium Mongolia Regel Extractive and its effects on CLA contents in digest tract, fat deposition of carcass and mutton flavor precursors in sheep were studied systematically in the thesis, which implements for synthesizing man-made Allium Mongolia Regel extractive.There are four parts in this study.Part1. Separation and identification of Allium Mongolia Regel extractive. Allium Mongolia Regel soluble-fat and soluble-water components in three different growth periods were extracted by instilling solvent methods. Fat-soluble extractive of Allium Mongolia Regel were analyzed by gas chromatogram-mass spectrometer, water-soluble extractive of Allium Mongolia Regel were analyzed by liquid chromatogram-mass spectrometer. The results indicated that main compound types of Allium Mongolia Regel soluble-fat extractive in three growth periods were basically identical, which main active materials were Diolefine, Ammonia, Spirits, Acids and some aromatic odor Phenols, but varieties of procreation prophase were most, that of procreation anaphase were least in three months. Main components of Allium Mongolia Regel soluble-water extractive in three periods were native compounds and ramifications. Mutual main active materials in three periods were Cysteine ramifications, Allixin, Benzopyran ramifications, Spirostane ramifications, Astragalin ramifications.Part2 Effects of Allium Mongolia Regel extractive on rumen ferment in sheep. Four healthy Mongolia wether sheep with permanent ruminal and proximal duodenum cannulas were selected, and fed diets on concentrate/roughage ratio of 30:70 and 40:60 respectively, to determine suitable addition of Allium Mongolia Regel extractive in vitro. The results showed that 335mg AWE/kg diets and 20mgAFE/kg diets were fit to ferment in concentrate/roughage ratio of 30:70, and 335mgAWE/kg diets and 40mgAFE/kg diets could promote fermentation in concentrate/roughage ratio of 40:60. Rumen fermentation in sheep by adding suitable AWE and AFE in concentrate/roughage ratio of 30:70 was studied. The results indicated that rumen NH3-N concentration and infusorian quantity were decreased significantly, and BCP content were increased significantly in Allium Mongolia Regel group and AWE group. Rumen liquid VFA concentration was markedly decreased in mixed group, therefore, which improved inner environment and promoted rumen fermentation.Part3 Effects of Allium Mongolia Regel extractive on CLA content of digestive tract in sheep.Four healthy Mongolia wether sheep with permanent ruminal and proximal duodenum were selected, and the diets of concentrate/roughage ratio of 30:70 were feed by sheep. Effects of Allium Mongolia Regel extractive on CLA content of digestive tract in sheep were studied by separating, purifying lactic acid bacteria of rumen and chyme in sheep and adding linoleic acid to cultivate. The results showed that lactic acid bacteria quantity was highest in mixed group both in rumen and duodenum, then in AWE group. CLA content was highest in mixed group, then in Allium Mongolia Regel group. Lactic acid bacteria quantity and CLA content in duodenum were more than those of rumen, producing CLA content by adding linoleic acid to cultivate lactic acid bacteria was more than that of rumen and duodenum by directing to measure, and lactic acid bacteria in rumen and duodenum belonged to Lactobacillus plantarum by 16SrRNA and traditional assay methodsPart4 Effects of Allium Mongolia Regel extractive on fat deposition of carcass and mutton flavor precursor in sheep.Sixty healthy Mongolia wether sheep with 15kg were randomly divided into five groups, which were control group, Allium Mongolia Regel group, AWE group, AFE group and mixed group(AWE and AFE) respectively. Each consisted of 12 sheep, which were three treatments and four repetitions. Diets of concentrate/roughage ratio of 40:60 were feed by sheep, and sheep were slaughtered in 15kg, 20kg, 30kg, 40kg body weight respectively. Effects of Allium Mongolia Regel extractive on fat deposition of carcass and mutton flavor precursor of carcass in different body weight sheep were studied. The results showed that fat deposition of subcutaneous fat, perinephric fat and total body fat were significantly decreased by decreasing FAS activity, increasing HSL activity and LPL activity in Allium Mongolia Regel and its extractive groups, and decreased SFA content of subcutaneous fat, longissimus muscle and perinephric fat and increased contents of MUFA and PUFA, which increased mutton quality and decreased odor of mutton. Fresh amino acids, IMP and reducing sugar contents in longissimus muscle and semimembranosus were increased by increasing contents of CP and EE in two profiles, which improved mutton flavor. |
