Zhi cao rheumatic and analgesic plaster is developed from the prescription of appropriate addition and subtraction component based on years of clinical experience of my tutor. It is composed of gardenia, kusnezoff Monkshood Root, safflower, Radix Gentianae macrophyllae and can dry yellow water,relieve the pain,detumesce,promote blood circulation. Mongolian literature and modern clinical practice has proved the sticking therapy has significant therapeutic effect on soft tissue injury, and has no toxic side effect, safe and reliable, simple and easy. This experiment observ the effect of Zhi cao rheumatic and analgesic plaster on AST1rats in different aspects and clarify its action mechanism on AST1.This paper consists of two parts, the first chapter is a summary, the understanding of soft tissue injury in traditional Mongolian medicine and research progress about the modern study of acute soft tissue injury progression and the composition of Zhi cao rheumatic and analgesic plaster. The second chapter is the part of the experiment, the experimental study of the effect of Zhi cao rheumatic and analgesic plaster on AST1rats.ObjectiveThe experiment observed the variation of the morphology, blood rheology, inflammatory factors and cytokines in rats before and after treatment after sticking Zhi cao rheumatic and analgesic plaster and clarified its action mechanism on AST1, improve the pasting Zhi cao rheumatic and analgesic plaster modern research experimental basis, provide the basis for further research and application.Method1.SD rats, male and female, a total of96, were randomly divided into6groups, normal group, model group, Zhi cao rheumatic and analgesic plaster high dose group, middle dose group, low dose group and Voltaren ointment group,16rats in each group, administrated after modeling24h, I times a day, a total of7times. The fourth day and eighth days, each soft tissue injury score the same phase sampling and observated the wound tissue morphology and infiltration of inflammatory cells under light microscope.2.Selected automatic blood rheology analyzer detect rats blood rheology.3.Using the ELISA method to measure the content of IL-1β,IL-6,PGE2,TNF-α.4.Immunohistochemistry was used to detect the expression of vascular endothelial growth factor cell injury rats (VEGF), basic fibroblast growth factor (bFGF) expression.Result1.Damage symptom index:Injury symptom score of Zhi cao rheumatic and analgesic plaster groups were lower than that of the model group. Histopathology:Pathological changes were significantly improved after administrating. Treatment group compared with the model group, there were significant differences except the low dose group.2.Indexes of blood rheology:After fourth days, compared with the model group, Zhi cao rheumatic and analgesic plaster high, middle and low dose group, the whole blood viscosity in high shear, low shear were significantly different (P<0.01; P<0.05). Zhi cao rheumatic and analgesic plaster in high, middle and low dose group, plasma viscosity and erythrocyte aggregation index were significantly different (P<0.01; P<0.05). After eighth days, compared with the model group, Zhi cao rheumatic and analgesic plaster high, middle dose group, the whole blood viscosity in high shear, low shear were significantly different (P<0.01), low dose group, the whole blood viscosity in high and middle shear had significant difference (P<0.05), low shear had no significant difference (P>0.05). Zhi cao rheumatic and analgesic plaster high, middle dose group, plasma viscosity and erythrocyte aggregation index was significant (P<0.01; P<0.05). Plasma viscosity and erythrocyte aggregation index of low dose group showed no significant difference (P>0.05).3.IL-1β content:Fourth days, eighth days the content of IL-1β in damaged tissue was significantly different among the Zhi cao rheumatic and analgesic plaster groups and model group (P<0.05; P<0.01).IL-6content:After fourth days, compared with the model group, the content of IL-6in Zhi cao rheumatic and analgesic plaster high dose was decreased (P<0.01), no significant difference in middle and low dose group (P>0.05). After eighth days, the content of IL-6in high and middle dose group of Zhi cao rheumatic and analgesic plaster was decreased(P<0.05, P<0.01), no significant difference in low dose group (P>0.05).PGE2content:The fourth and eighth days, the content of PGE2in damaged tissue was significantly different among the Zhi cao rheumatic and analgesic plaster groups and model group (P <0.05; P<0.01).TNF-a content:The fourth and eighth days, the content of TNF-a in damaged tissue was significantly different among the Zhi cao rheumatic and analgesic plaster groups and model group (P <0.05; P<0.01).4.The average optical density of VEGF and bFGF values, have decreased in different degree. Except the exception of small dose group VEGF optical density value, other groups were compared with the model group were significantly different, but bFGF optical density values for each group compared with the model group were significantly different.Conclusion1.Zhi cao rheumatic and analgesic plaster can obviously reduce the local swelling, blood stasis of acute soft tissue injury. And also reduce the damage infiltrating surrounding matter and blood neutrophils local muscle fiber.2.Zhi cao rheumatic and analgesic plaster can reduce the blood viscosity in rats of acute soft tissue injury, improve red cell membrane function, improve microcirculation, improve blood hypercoagulable state, high viscosity.3.Zhi cao rheumatic and analgesic plaster can significantly reduce the rat model of acute soft tissue injury in local tissue inflammatory cytokines of IL-1β,IL-6, PGE2, TNF-α.4.Zhi cao rheumatic and analgesic plaster can reduce the expression of injury in a rat model of local tissue damage in VEGF and bFGF of acute soft tissue.5.Zhi cao rheumatic and analgesic plaster can significantly shorten the course of soft tissue injury, promoting repair of injured muscle fibers, have a good effect on ASTI. Mechanism of Zhi cao rheumatic and analgesic plaster in the treatment of ASTI perhaps is improving the microcirculation, changing a hypercoagulable state.high viscosity.reducing the local tissue damage and inflammatory cytokines in IL-1β, IL-6, PGE2, TNF-α content, and decreasing VEGF and bFGF expression.
