Objective:This experiment was designed to establish a rat ischemic syndrome of Yang deficiency and blood stasis model.To explore the soft (liquid) capsule with the function of warming yang and activating blood with ischemic syndrome of Yang deficiency and blood stasis rat modelon serum markers of platelet activation GMP-140 content,receptors and ligands GPâ…¡b/â…¢a vWF concentration,TXB2 and 6-Keto-PGF1 α content and platelet ultrastructure affect aggregation rate which revealed the effect that the soft (liquid) capsule with the function of warming yang and activating blood could prevent and treat ischemic heart disease of Yang deficiency and blood stasis model to provide a scientific theory for clinical practice.Materials and methods:Use high-fat diet,vasopressin (Pit) subcutaneous injection and cold stimulation method to establish ischemic heart disease with yang deficiency and blood stasis rat model.Apply advanced methods such as physical and chemical examination of color Doppler ultrasound,cardiac electrophysiological recordings Miriam, enzyme-linked immunosorbent assay, biochemical detection,optical microscopy and other modern detection techniques.to evaluate the validation of model establishment and to detect GP â…¡b/â…¢a serum concentration,vWF concentration,GMP-140 content,TXB2 and structure of 6-Keto-PGF1 α content.platelet aggregation and platelet electron microscope.Compare the differentiation of GP â…¡ b/ â…¢ a serum concentration,vWF concentration,GMP-140 content,TXB2 and structure of 6-Keto-PGF1 α content.platelet aggregation and platelet electron micros cope among all the groups.All the results was used to assess the function of platelet and morphology changes to provide more accurate and objective indicators and explorethe mechanism of soft (liquid) capsule with the function of warming yang and activating blood in the prevention of ischemic heart disease Yang stasis syndrome.Results:1. Compared with normal rats,the ischemic heart disease in experimental model group, Electrocardiogram blood lipids, enzymes, HE staining of myocardialwere significant differences (p<0.05), prove the model of myocardial ischemic changes;Rats clammy skin, the temperature drops, apathetic, lying curled get together, curled up less dynamic, lip weeks cyanosis, dull red, loose stools thin, such as performance, and GMP-140 content in serum of rats, GPâ…¡b/â…¢a and vWF concentration and TXB2/ 6-Keto-PGFlα ratio showed elevated platelet aggregation occurs activation, and platelet aggregation rate increases, the electron microscope platelet aggregation height change, proving to be successful Yang deficiency and blood stasis rat model.2. Compared with model rats, the serum levels of GP-â…¡bâ…¢ã€vWFã€GMP-140〠TXB2/6-Keto-PGF1α were significantly reduced in warming yang and activating blood group (p<0.01), and the serum levels of the platelet aggregation was significantly reduced(p<0.05), and improved the platelet ultrastructur in warming yang and activating blood group.Conclusion:Soft (liquid) capsule with the function of warming yang and activating blood might reduce a granule membrane protein GMP-140 content,inhibit platelet membrane glycoprotein GPâ…¡ b/â…¢a aggregation,reduced and reversed the process of platelet activation,inhibit platelet adhesion, reduced vWF expression,reduce platelet aggregation and release reaction,reduce TXB2 levels and increase 6-Keto-PGF1 α levels to correct TXB2/6-Keto-PGF1 α imbalance.further affect the signal platelet function,inhibit platelet aggregation and activation, revers ischemic heart disease with blood stasis and deficiency of yang rats model changes,lowered blood hypercoagulable state.Thus inhibiting endothelial injury at thrombosis and achieve the purpose of the prevention and treatment of ischemic heart disease.
