Objective Polycystic ovary syndrome is a common gynecological endocrine disease characterized by Polycystic ovary-like change, anovulation, hyperandrogenism and insulin resistance, main clinical manifestations of hirsutism, acne, menstrual scarce, and not being pregnant. Approximately, 5%-10% of the gestational age women is involved with PCOS. The cause and mechanism of pcos is complex and unclear, such as many abnormal changes of neuroendocrine, sugar, fat and protein metabolism. These can affect seriously women’s physical and mental health.With the change of life and environment, the incidence rate has increased gradually for years. Because of the side effects and recurrence rate from traditional medicine, surgery, so it is very important to search for new treatments. Traditional Chinese medicine in the treatment of PCOS is unique to regulate endocrine restore menstrual cycles, improve the ovulation rate and pregnancy rate,and reduce the recurrence rate. In the present study,we research the effects and mechanism of Dingkun Dan on PCOS rat model, It is designed to provide an experimental basis for Ding Kun Dan in the clinical treatment of PCOS.Methods The present study is divided into four parts:At first, the effect of Dingkun Dan on hormone and insulin, Leptin with the PCOS rats. This part presents the influences of Dingkun Dan on T, LH, FSH and RI and leptin in peripheral blood including exploring Dingkun Dan on the hypothalamus- pituitary- ovarian axis and the regulation of glucose and lipid metabolism. Next, the influence of ovarian morphology on PCOS rats given Ding Kun Dan is discussed. In order to understand ovulation induction, the ovarian morphology is observed using the optical microscopy. Thirdly, the effect of Angiogenesis-related factors is carried out. In recent years, around angiogenesis theory, the treatment mechanism of Ding Kun Dan on rats model with pcos is investigated. At last, we research the endometrial receptivity associated markers of PCOS model rats given Ding Kun Dan,to probe the treatment mechanism in terms of endometrial receptivity. A PCOS model was established using testosterone propionate injection combined with humen chorinic gonadotropin(HCG). After the successful construction of the model, Rats were divided into 4 groups: model control group, Ding Kun Dan group, Clomiphene group and normal control group. We compared ovarian morphology, sex hormones,insulin and leptin levels in rats, the expression of vascular endothelial growth factor(VEGF) in ovary and homeobox gene 10(HOXA10) in uterus before and after dosing.Results In a comparison to the model group, Ding Kun Dan could promote follicle, and increase the number of corpus luteum. Additionally, the serum Luteinizing hormone(LH), androgens(T),Insulin and leptin level of the Ding Kun Dan group declined, but follicle stimulating hormone(FSH) increased. The expression of VEGF in ovary reduced while HOXA10 in uterine rised. On top of that, their differences happened were statistically significant with P<0.05.Conclusion Ding Kun Dan had an ovulation function and could increase the role of endometrial receptivity. The mechanism might relate to the regulation of the serum sex hormones, Insulin and leptin levels, the decrease of VEGF in ovary, and the addition of HOXA10 in uterus in PCOS model rats.
