The Comparative Study on Neuron and Astrocyte Cytotoxity from Serumand Cerebrospinal FIuid by Chinese Materia MedicaObJective The in vitro experiment of pharmacodynamics on Chinese materia medicausuaIIy empIoys the method of serum pharmacoIogy, however which might not adapt inthe ceIIs behind the bIood-brain barrier TO expIore an useful method by which thepharmacoIogicaI thects of Chinese materia medica on the CNS could be correctlyinvestigated, the cytotoxity of both serum and cerebraI-spinaI nuid (CSF) was observed.thethods CSF and serum of the rabbit were taken affer administration of Chineseherbal compound (CHC) QingNaoYiZhi and added into the medium of cuItured rat corticafneuron and astrocyte in proportion of 5%, 10% and 20%. Typan bIue staining, lactatedehydrogenase (LDH) mensuration and Mrr test were perfOrmed to observe the moFtaIityand activity of the above tWo kinds of ceIl.ResuIts For the neuron and astrocyte, the serum group and the CHC-serum groupmanif6sted the higher cell death rate, the more LDH cont6nt and the fower OD vaIue thanthe controI group, furthermore, there is no difference betW6en those tWo groups. It meantthat either serum or CHC-serum could produce the Cytotoxity on the neuron and astrocyte,which seemed to concem with the serum itseIf, but not the CHC. ln addition, the CSFgroup and CHC-CSF group showed non-cytotoxop on the neuron and astrOcyte.Concluslon Since the serum braught the great cytotoxity to the ceIls behind thebIoad-brain barrier, itS toxity wilI cover the drug effeCt if the serum pharmacoIogy isadopted to observe the pharmacodymamics to the CNS. Howevet. it was deemed that the'Cerebrospinal FIuid PharmacoIogy of Chinese Mat6ria Medica" is worth fOr further study...
