BACKGROUNDThe mechanism of secretion and regulation of gastric acid is an important subject in basic research of gastroenterology. There is a tight relation between occurrence of ulcer and secretion of acid. Gastric acid is secreted through parietal cells. Parietal cells could secret HCI contradictorily to the stairs of concentration of hydrogen ion. It's complex work to regulate the secretion of HCI of parietal cells. It's very useful to study the function of parietal cells and to reveal the physiological mechanism of HCI secretion through isolation of parietal cells. The potassium ion channel in parietal cells could be studied through patch clamp technique in the level of a single cell and its membrane. The location, classification and characteristic could be elucidated. The physiological, pathological or pharmacological action of potassium channel in parietal cells could be further researched. Interleukin Ifl is the most intense immunological factor which has the effect of protection, anti-ulcer, anti-secretion and which postpone the empty of stomach. There are receptors of IL-1?which could combine with EL 13 and then generate the restraining effect to secretion of gastric acid of parietal cells.AIMS(1)To investigate the effect of environmental hyperthermia on gastrin, somatostatin and motilin in rat's ulcer antral mucosa. (2) Revised immunoelectron microscopic technique was used to detect and to analyze the G cells and D cells of rat's antral mucosa. (3)To establish the scrape method for acute isolation of rat parietal cells and to identify the parietal cells. (4)To study, the characteristic of potassium channel or rat parietal cells throughpatch clamp technique. (5)To study the effect of IL 1B to parietal cells in the level of membrane and to study the regulation effect of EL 1B to potassium channel of parietal cells.METHODS(1)Forty two Wistar rats were divided into 6 groups. The acetic acid gastric ulcer model was induced successfully. The level of gastrin, somatostatin and motilin in rat's antral mucosa was measured with radioimmunoassay method. (2) Revised immunoelectron microscopic technique was used to detect the G cells and D cells of rat's antral mucosa. The number of colloidal gold in G cells and in D cells was analysed through quantitative analysis. (3)Acute isolation of rat gastric mucosal cells was performed through scrape method and through protease and collagenase digestion. Parietal cells were observed and identified under a phase-contrast microscope, through hematoxylin and eosin staining, under an electron microscope, under a fluorescence microscope, and through immunocytochemistry technique. The diameters of parietal cells were counted. (4)Make the electrophysiological record of potassium channel of rat parietal cells through whole cell's patch clamp technique and analyze the basal characteristic of channel. (5)Observe the effect of IL 1B to parietal cells through hematoxylin and eosin staining and through immunocytochemistry technique. Record and analyze the effect of IL 1B to potassium channel of parietal cells through whole cell's patch clamp.RESULTS(1)in gastric ulcer rat the level of gastrin and motilin in antral mucosa increases, that of somatostatin declines. The level of gastrin and motilin in antral mucosa hi the high temperature ulcer group increases less than that in the normal temperature ulcer group. The level of somatostatin in the high temperature ulcer group declines less than that in the normal temperature ulcer group. (2)The inununological granules of colloidal gold distribute in G cells and in D cells. Gastrin labeled golden granules or somatostatin labeledones present mainly as lobation-like or island-like congeries. Most of the golden congeries dissociated in cytoplasm of G cells or D cells, being in cells' ground membrane side. A few of the golden congeries locate in nuclear.
