The research project is a part of the national "973" Project:"Basic Research of the Key Problem of Chinese Herb Prescription-Research of the Prescription to Treat Internal and External ZHENG". The research object is DCXW. The clinic use of DCXW is to treat migraine. The research content is under the guide of TCM theory. First,use the modern pharmacokinetics research thoughtway and method to screen the effect part of DCXW and carry out its combinatorial chemical research. The pharmacokinetics research chooses effective ingredients Ferulic acid and Gastrodin as the research object Analysis the ingredients in and out blood after taken DCXW by LC-MS. Restrain thrombocyte to release 5-HT and prevent Ca channel 'of thrombocyte experimen are chosen to carry out the pharmacodynamics research. Also analysis the PK-PD model.The result of the research:(1) The effective part of Lisgusticum chuanxiong Hort and Gastrodia elata Bl. in DCXW is extract of 95% alcohol. Lisgusticum chuanxiong Hort. can restrain thrombocyte to release 5-HT,so it can reduce the vein fit caused by 5-HT. Gastrodia elata Bl. can through the "exhaust mechanism" to reduce 5-HT. (2) Ferulic acid can be absorbed very rapidly through stomach after taken DCXW,the distribution and elimination is fast,too. So it can relax the symptom of migraine quickly. The absorb speed of Gastrodin is a little slowly. The distribution is fast,but the elimination is very slowly. It can maintain theeffect of DCXW. The distribution of Ferulic acid and AUC is bigger than Gastrodin. (3)The results of LC-MS show that the ingredients in blood is limited after taken DCXW,they can be determine the nature and quantity. Gastrodin has a tide combination with plasma albumen. (4) Through PK-PD analysis,we find the pharmacology effect of DCXW is lag to the concentration of Ferulic acid and Gastrodin in blood. It has a good relativity between the pharmacology effect and the concentration of effect compartment The relationship of effect and concentration of effect compartment accords to the Emax model.
