The Study on Bio-ethnopharmacuetic analytical pharmacology was put forward by Professor Huang Xi that according to the themry of Traditional Chinese Medicine. This research aims was to discovering biologically active compounds with high probability in traditional Chinese herb remedies was performed from a pharmacokinetic. It includes pharmacokinetics and pharmacodynamics research of recipe. In present study,we applied high performance liquid chromatography (HPLC) to determine Ferulic acid(FA) and Gastrodin(Gas) that derived from extract of Da-Chuanxiong-Wan (DCXW) and reseached pharmacokinetics of them.DCXW is made up of ligusticum wallichii and tall gastrodia tuber and commonly used for the treatment of migraine diseases.If Ferulic acid(FA) and Gastrodin(Gas) could be absorbed into the body and induction effects of parent recipe, these compounds could be the basic ingredients of pharmacodynamics of recipe.Aim:The objective of our present studies was to estabilish the methode to determine Ferulic acid(FA) and Gastrodin (Gas) by HPLC, and to study the9pharmocokinetic of FA in serum of healthy rabbits and Gas in plasma of rats after oral administration of extract of DCXW. The analytical method was adopted high-performance liquid chromatography (HPLC) and analysis the ingredients of DCXW in vivo and in vitro by LC-MS. Method:1. By using the developed high-performance liquid chromatographic (HPLC) method which was previously established in our laboratory, we have determined Ferulic acid(FA) in serum of rabbits after oral administration of the extract of DCXW: The serum of rabbits contained Ferulic acid and internal standard coumarin (IS) were firstly treated in boiling water-bath and then used methanol to deproteinate protein,and then blowed methanol by nitrogen gas,at last was novo-lysis by methanol. Methanol-water-aceticacid (49 '. 50.745 ' 0.255, v/v)was used as the mobile phase,while the Inertsil ODS-3 (4.6 ?250 mm, 5 um) as a fixed phase.The fluid flowing rate was 1 .Oml/min, FA was detected at the UV wavelength of 322nm.In this research, we have collected serum samples of rabbits after oral administration of DCXW at a dosage of 11.667 g/kg to healthy rabbits at different times,and determined Ferulic acid(FA) in serum samples treated by methode we estabilished by HPLC.The pharmacokinetic parameters were analysed by 3p97 software.2. In this research,we established a new method to determine Gastrodin (Gas) in plasma of rats after oral administration of extract of DCXW by RP-HPLC: The plasma samples of rats contained Gastrodin and internal standard triogyl was firstly treated by ethanol to deposit protein in plasma, and then blowed ethanol by distilled water, acetonitrile- alcahol water-aceticacid (2.64:97.36:0.2, v/v)was used as the mobile phase,while the Diamonsil (TM)(C18 5 u m , 4.6?150 mm) as a fixed phase.The fluid flowing rate was 1.0 ml/min, Gas was detected at the UV wavelength of 221 nm.We have collected plasma samples of rats after oral administration of DCXW at a dosage of 11.66710g/kg to healthy rats at different times,and determined Gastrodin (Gas) in plasma treated by methode we established by RP-HPLC.The pharmacokinetic parameters were analysed by 3p97 software.3.In the couse of research we analyised ingredients of DCXW by LC-MS.Its aim to search the basic ingredients of pharmacodynamics of DCXW that could be remission the symptom of the migraine.?The ingredients of DCXW absorbed in vivo after oral administration by rabbits and in vitro were determined by LC-MS, Methanol- water-aceticacid(40:59.7:0.3 , v/v) was used as the mobile phase,while the Hypersil ODS column (4.6 X 10 mm 5 urn) as a fixed phase.The fluid flowing rate was l.0 ml/min, FA was detected at the UV wavelength of 322 nm.
