Studies Of Fructus Jujubae Polysaccharide (FJP) On Replenishing Qi And Nourishing Blood

Objective: To reveal the mechanism of Fructus jujubae polysaccharide(FJP) on replenishing qi and nourishing blood, we studied the effects of FJP on the immunosuppressive mice model and both qi and blood deficiency mice or rat model, and at the same time, established a new both qi and blood deficiency animal model.Method: We study systematically chemical property of experimental FJP. Adopting the immunosuppressive mice model induced by injection of cyclopho sphamidc(CY) as researching object, we observed the effects of FJP on immune system according to the follow indexes, such as phagocytosis function of celiac phagocyte, the formation of hemolysin and hemolysin plaque of serum, the transformation rate of lympholyte and the ratio of peripheral T-lymphocte, the level and activity of IL-1 and IL-2 produced respectively by celiac phagocytes and spleen cells, the level of SIL-2R in serum, the reproduction of spleen cell stimulated by ConA and LPS in vitro, etc(replenishing qi). Adopting both qi and blood deficiency mice or rat model induced by blood releasing and injections of CY as researching objects, we observed the effects of FJP on hematopoietic system, immune system and the energy metabolism according to the follow indexes, such as the hemogram, the tissue slice and ultrastructure of spleen and thymus, the level of IL-2, IL-6,EPO and GM-CSF in serum, the activity of the Na+-K+-ATPase, Ca++-ATPase, Mg++-ATPase, Ca++-Mg++-ATPase in erythrocyte ,and the relative gene expression of cytokine in thymus, spleen and bone marrow etc(replenishing qi and nourishing blood).Result:Chemical study of polysaccharide: we selecte the FJP sedimentated by 60% ethanol as the experimental sample, which is made up of six components. Two of these components can't be studied deeply because of rareness, the amount of the molecule weight and molar ratio of the others are list as follows: ZJ-1, 9848, Glucose: Galactose: Arabinose = 1: 2: 5; ZJ-3, 8991, Glucose:Xylopyranose:Arabinose=11.53:15.9:13;ZJ-4,4694,Glucose:Xylopyranose:Arabinose=2.4:9:1;ZJ-6,4206,Galacturonicacid:Glucose: Xylopyranose: Arabinose=100:6.27:1.3:0.489.Effect on immunosuppressive mice model: FJP can promote remarkably the rate and index of phagocytosis of celiac phagocyte in immunosuppressive mice , advance obviously the formation of hemolysine and hemolysim plaque ,and increase thetransformation rate of lymphocyte and the ratio of the peripheral T-lymphocyte(p<0.01).FJP can increase obviously the activity and formation of IL-1 and IL-2 produced respectively by celiac phagocytes and spleen cells, depress the level of SIL-2R in serum ,and advance the reproduction of spleen in vitro stimulated by ConA and LPS also(p<0.01) .Effect on both qi and blood deficiency animal model: FJP can promote remarkably the hemogram level of both qi and blood deficiency mice model induced by blood releasing and injections of CY , increase the thickness of thymic cortex, the quantity of cortical lymphocyte and the spleen lymphocyte ,enlarge splenic lymphatic nodules(p<0.01) , promote the GM-CSF level in serum(p<0.01).FJP promote remarkably the hemogram level at 5th,9th,14th day of both qi and blood deficiency rats model induced by blood releasing and injections of CY(p<0.01), increase the thickness of thymus cortex, the quantity of cortical lymphocyte and the spleen lymphocyte, enlarge splenic lymphatic nodules(p<0.01), promote remarkably the levels of IL-2, IL-6,EPO in serum(p<0.01), enhance the level and activity of Na+-K+-ATPase, Ca++-ATPase, Mg++-ATPase, Ca++-Mg++-ATPase in erythrocyte (p<0.01) . FJP make the body-density of mitochondria and euchromatin, and Sm increase obviously , the body-density of heterochromatin , S and Nucleus S decrease obviously(p<0.01).
