| OBJECTIVE: ET was discovered that it relat with microcirculation of pancreateas in acute pancreatitis in recently years, though bad microcirculation of pancreateas would increase the unbanlance of pathnicphysiology and morbility. The experiment aids to measure the concentration of ET which changed in ANP in rats by radiaimmunology and Immunohistochemistry , RT-PCR and so on. METHODS: 1 Animal to divide into groups. One hundredred and eight rats was divided into four groups: included sham operation(group A), artificial AP group(group B) , dopamine treating group by arterial injection(group C) and dopamine treating group by tail vein injection(group D). 2 Model: The common bile duct of rats were injected to reverse 4.5% sodium taurocholate. 3 Measure of blood samples of all groups by radiaimmunology. 4 Dyeing of pancreas tissue by immunohistochemistry. 5 Preparation of total RNA of pancreas. 6 The reaction of RT-PCR of mRNA of ET-3. 7 RNA and the product of RT-PCR reaction agrose electrophoresis. 8 Pathology mark of pancreas: according Kussake' meathods give a mark for inflammation and edema, hemorrhage ofpancreas tissue. RESULTS: 1 radiaimmunology: ET in artificial group increase obviously than in other groups at 1h , 6h , 24h. 2 Immunohistochemistry: photodensity in artificial group significance increas than in other groups. 3 Preperation of total RNA of pancreas: the ratio of extracted RNA was among 1.8~2.0. the result accord with demands of RT-PCR reaction. 4 Semi-quantity RT-PCR of ET-3 mRNA: the express of ET-3 mRNA in all groups at 6 h were the highest than at 1 and 24 h. 5 pathology mark: there were low score in group C than in group D. CONCLUSION: the levele of ET in portal vein could make known the extent and degree of pathological changs in pancreas, there were more effect dopamin treating by arterial injection than by tail vein injection. Express of ET-3 mRNA was according to the degree of pathological changs in ANP.
