Objective: To investigate the change of intestinal motility after abdominal surgical manipulation and the modulation effect of KangYanLing. Method: Ten of 70 SD rat were selected randomly asnormal controls, the rest were designed to operation group and KangYanLing group and there were 30 rats in each group. Animals of operation group received the abdominal manipulation procedure, while that of KangYanLing group were administrated kangYanLing by gastric tube before surgery. Gastrointestinal transit was measured in control and experimental animals by evaluating the intestinal location of fluorescence marker. The mechanical activity was measured by multichannel Biopac system. The change of contractile response and [Ca2+]i of small intestinal smooth muscle cell induced by Ach were quantified. Result: Surgical manipulation caused a significant delay of the in vivo intestinal transit and decrease in the contractile activity of small intestinal circular muscularis strips. The contractile response and increase of [Ca2+]i induced by Ach were suppressed by surgical manipulation. KangYanLing averted small intestinal smooth muscle dysfunction. Conclusion : The data demonstrate that surgical manipulation can result in small intestinal smooth muscle dysfunction by inhibiting the increase of [Ca2+]i, the administration of kangYanLing before operation may prevent surgery-induced ileus through the effect on the [Ca2+]i, and/or the Ca2+ sensitizition.
