ObjectiveTo study semi bionic extraction technology of Angelica sinensis and Astragalus membranaceus, and to confirm effects of semi bionic extraction of A. sinensis and A. membranaceus(SEA) on gastrointestinal motility dysfunction in abdominal surgery rats, and to explore its mechanisms, the experiment was arranged. MethodsBy using uniform design, the best extraction process of SEA was found based on pharmacodynamics method of enriching blood and benefiting qi effects of A. sinensis and A. membranaceus. SD rats were fasted for 24 h, General anesthesia was operated by chloral hydrate, The abdominal cavity was opened, Two transverse incisions were operated at olon appendix 2cm distance(2cm apart from the incision), incision suture in situ, then peritoneal suture, muscle layer and skin, So the abdominal operation model was copied in rats. After operation at 24 h, the rats were randomly divided into operation model group, positive control group, AEA control group, high, medium and low dose group of SEA, sham operation group,(only open the abdominal cavity, and immediate closure), 8 rats in each group; sham operation group and the model group was gavaged with normal saline by 10 m L/kg, the positive control group(3.5 mg/kg of cisapride), AEA control group(5.2g/kg of SEA), high, medium and low dose group(10.5, 5.2, 2.6g/kg of SEA). Administration was operated at 24 h after operation, continuous 10 d. During the experiment, the general state of the rats were observed, and the time of first defecation and stool weight in 6h were recorded after the first 3d administrated, food intake and gastrointestinal propulsive index were determinated also; the levels of MTL in plasma and GAS in serum were determinated by ELISA; H-E staining to colon tissue, morphology were observed under microscope, the ultrastructure of colonic ICC were observed under electron microscope; the content of myosin light chain kinase(MLCK), actin and myosin were detected immunohistochemistry. Results1) The optimum extraction process of SEA as follows: the amount of artificial gastric juice is 8.0 times as the number of medicines for first extraction, and extraction time 3.0h; the amount of the artificial intestinal juice 7 times for second times extraction, and extraction time 4.0h.2) Compared with the sham operation group, varying degrees of spirit was dispirited, slow, defecation quantity is little, weight loss and etc in model rats; At 9d after the administration, gastrointestinal propulsion index decreased, the levels MTL in plasma and GAS in serum were significantly reduced, gap of colonic smooth muscle cells become larger, cytosolic was decreased, vacuolar changes was significantly, vascular smooth muscle was markedly dilated, thinning, muscle fibrosis was visible, granulation, enteritis fat particles is abound, the ultrastructure of ICC was obviously damaged, a large number of mitochondria rupture was dissoluted; the contents of MLCK and actin were significantly decreased.Compared with the operation model group, the body weight of rats were significantly increased in AEA control group and high, medium, low dose of SEA group, the first defecation time was obviously shortened in the positive control group, high or medium dose of SEA group, the amount of defecation in 6h was significantly increased, and food intake increased during 24 hour; At 9d after the administration, gastrointestinal propulsion index and the content of MTL in plasma and GAS in serum increased significantly in the positive group, AEA group,and high or medium dose of SEA group. colonic pathological changes was improved and the number of interstitial ICC were significantly increased in the positive group, AEA group,and high or medium dose of SEA group. Moreover, the content of MLCK, actin and myosin were relatively increased in different degree in the positive group, AEA group,and high or medium dose of SEA group. ConclusionsThe extraction with nourishing blood and benefiting qi can obtain by using semi bionic extraction technology in Angelica sinensis and Astragalus membranaceus. The certain therapeutic effect of SEA on gastrointestinal motor dysfunction were determinated in abdominal operation rats, The mechanisms associated with the levels of MTL and GAS in blood, the contents of MLCK and actin in colon were increased.
