Insulin resistance (IR) is the chief feature of type 2 diabetes mellitus. IR decreases the insulin biological effect of human body in the target organism. The body has to secrete more insulin to overcome IR.IR will occur at pre-receptor ,receptor or post-receptor sites. Since the theory of diabetes millipidus has been proposed, the role of dyslipidemia in the pathogenesis of type 2 DM is emphasized. And the relationship between free fatty acid (FFA) and insulin resistance (IR) has been paid more attention. But the mechanism of IR induced by FFA is not clear. The purpose of this study is to investigate the prevention of IR and its possible mechanism by increasing dietary polyunsaturated free fatty acids (PUFAs) in the insulin resistance rats induced by high-fat diet in a equal calorie intake.1. The study on the prevention of IR of polyunsaturated free fatty acids (PUFAs) in the ratsinduced by high-fat dietObjective : To investigate whether or not PUFAs have the prevention effect of IR hi the rats induced by high-fat diet.Methods: There were 75 male Wistar rats including control, high fat-fed group, pioglitazone group , n-3 fatty acids group and n-6 fatty acids group in the study. The high-fat-fed group was fed a diet containing 58.8% saturated free fatty acids. The n-3 fatty acids group was also high-fat diet but 20% of sature fatty acid was replaced with EPA-E and DHA-E. The n-6 fatty acids group was high-fat diet but 20% of sature fatty acid was replaced with C 18:2 in soybean. The content of C 18:2 in soybean was measured by GC-MS method . The pioglitazone group was administered with 10mg/kg/day of pioglitazone by gastrogavage,and also the rats were fed high-fat diet. Each group rats were was killed at week 11.The dietary amount and the calorie were checked up every day. The body weight, the content of visceral fat ,fasting blood glucose, non-fasting blood glucose ,30,60 and 120min blood glucoseafter oral glucose intake and the glucose area under curve, 40, 90 and 120min blood glucose after insulin injection subcutaneously and the glucose area under curve at the insulin tolerance, serum insulin , insulin resistance index, serum cholesterol and serum triglyceride were assayed.Results :There was no obviously difference of the dietary amount daily among 5 groups. There were no difference of dietary calorie intake daily among 4 groups except for normal group. The dietary calorie intake was higher among 4 groups was higher than normal group.The body weight from week 4 to week 10, fasting blood glucose ,30, 60 and 120min blood glucose after oral glucose intake and the glucose area under curve, 40, 90 and 120min blood glucose after insulin injection subcutaneously and the glucose area under curve at the insulin tolerance, serum insulin , insulin resistance index, serum cholesterol and serum triglyceride were significantly higher in high fat-fed group than that of the control. The others such as non-fasting blood glucose, HDL and LDL levels were not obviously difference compared with the control.The body weight from week 2 to week 4 was higher in pioglitazone group than that of high-fat-fed group. But its body weight in the other weeks had not been effected. The body weight from week 1 to week 10 was higher than that of normal group. The content of visceral fat, fasting blood glucose , 30, 60 and 120min blood glucose after oral glucose intake and the glucose area under curve, 40,90 and 120min blood glucose after insulin injection subcutaneously and the glucose area under curve at the insulin tolerance, serum insulin , insulin resistance index, serum cholesterol and serum triglyceride were significantly lower in pioglitazone group than that of the high-fat-fed group .In n-3 fatty acids group, the body weight from week 1 to 10 , the content of visceral fat, fasting blood glucose , 30 and 120min blood glucose after oral glucose intake and the glucose area under curve, 40min blood glucose after insulin injection subcutaneously and the glucose area under curve at the insulin tolerance serum insulin , ins...
