Objective: To investigate the frenqency of variant at Mspl sites of apoAI gene and relations to coronary heart disease (CHD) and serum lipid levels in Chinese population. Methods: The restriction fragment polymorphisms(RFLP) at two Mspl sites in the 5' end at -75bp and +83bp of apolipoprotein AI gene were detected using PCR in 240 coronary heart disease patients and 196 healthy subjects from a population of Chinese Hans in Tianjin. Results: Both in CHD group and Control group, G/G genotype was the most frequent one, but the frequency of A/A genotype in CHD group was significantly lower than that in control group, p<0.05. No differences were found in the frequencies of rare A allele for -75bp and rare T allele for +83bp. In the CHD group, male subjects with A/A genotype had a lower serum mean concentration of HDL compared with male subjects with the genotype G/G, p< 0.05. male subjects with T allele had a lower serum mean concentration of HDL compared with male subjects with the genotype C/C, p<0.05. Conclusions: These results suggest that Mspl RFLP at -75bp of apoAI gene is associated with coronary heart disease to some extent in Chinese population. In the CHD group, the A/A genotype or T allele of apoAI gene is associated with the decrease of serum HDL level.
