Benign prostatic hyperplasia (BPH) is a common disease in male adult and old men it severely influences the quality of life, and so physical and mental health. The incidence of the disease is in creasing year and year along with coming of long-life society in our country. Up to now, the cause and pathogenesis of the disease are completely clear. Based on the changes of weights, morphologic and the rate of proliferation and apoptosis, the expression of growth factors, genes, estrogen and androgen receptors, this study researched the mechanisms affected the growth of mice prostate by epidermal growth factor, estrogen and androgen, by with it explores the etiology of BPH. The results of the study are as follows:1. The research of the mechanism affected the growth of mice prostate by epidermal growth factorWe first observed the effects of EGF on weight and morphology of the prostate, based on it, the expression of ER,AR,bcl-2,bax,c-myc,EGF,EGF-R,bFGF, the changes in proliferation and apoptosis rates of prostatic cells under the effects of EGF were determined to study the effect and its mechanism of EGF on the prostate of Kunming male mice.Sixty Kunming male mice (aged from 22-27 days, mean 24.7 days; weighting 15-20g, mean 16.8g; certificate: YDZ04056) were divided randomly into 3 groups: A,B,control group, each twenty (1)A: subcutaneously injected EGF diluted by distilled water for 28 days, 2μg/d for one mice; (2)B: subcutaneously injected EGF diluted by distilled water for 28 days, 1μg/d for one mice; (3) control group: subcutaneously injected distilled water. For above 3 groups, killed them on 28th day, took out the prostate and observed below items: the weight and morphology of prostate, the expression of ER and AR, the rate of proliferation and apoptosis, the expression of bcl-2,bax,c-myc, and the expression of EGF,EGF-R,bFGF. Weighed their prostates by analysis balance, observed the changes of morphologic, analyzed the others numerically by flow cytometry (FCM). The expression of above mentioned receptors, genes and growth factors were presented with labeled rate ( percentage, %) and expression level ( presented by fluorescence channel numbers collected as linear data). Rates of apoptosis and proliferation were expressed by percentage (%) of apoptosis and proliferation. All data were expressed with X±SD for statistics treatment.1.1 Change in weight of the prostate A group 59.400±8.750mg,B group 38.200±4.320mg, control group 22.100±4.960mg,there was significant difference in different group(P<0.01).1.2 Morphologic changes no significant difference between A and B group, glandular epithelia showed significant hyperplastic changes and the hyperplastic epithelia formed dense plicate and papillary structures filling the lumina of the prostate glands.1.3 Expression of ER and AR Expression of ER (1)The labeled rate: A group 34.850±8.800,B group25.525±6.782,control group 17.638±5.877,there was significant difference between experiment group and control group(P<0.01),but no significant difference between A and B group(P>0.05); (2)The expression level : A group 5.204±0.160,B group 4.713±0.353,control group 4.175±0.101,there was significant difference in different group(P<0.01).Expression of AR (1)The labeled rate: A group 35.012±6.292,B group 33.612±10.589,control group 21.712±2.921,there was significant difference between experiment group and control group(P<0.05), but no significant difference between A and B group(P>0.05);(2)The expression level : A group 6.433±0.488,B group 5.990±0.427,control group 5.032±0.229,experiment group was higher than control group(P<0.01),but no significant difference between A and B group(P>0.05).1.4 Changes in rates of apoptosis and proliferation and expression of genes Changes in rates of proliferation A group35.625±6.968,B group 30.375±3.777,control group26.309±2.020,there was significant difference between A group and control group(P<0.01),but no significant difference betwe...
