Endometriosis is a kind of common diseases among childbearing age women, however it seens to difficult to be overcome up to now. In the recent years, incidence of the endometriosis sends up rising trend, and the patients occupied about 30-40% of gyneacology outpatient service, patients needed the surgical operation to cure 20-30% of gynecology surgical operation. The etiological mechenism for the endometriosis could be complicated, and it may be still not very clear at present, so also haven't ideal therapeutic effect. This subject draws up to observed by the electron microscope, so as to find ectopic leisions of endometriosis and measure some connelated genes in the blood and the ectopic leisions among the patients of Xinjiang Uygurs and Hans nationality. Through cultivating the normal endometrial cells, eutopic and ectopic endometrial cells of the endometriosis patients outsite bodyand measuring matrix metalloproteinase-2 ( MMP-2) level and the changes of this enzyme standards after some drugs' effects, It can be initially explored possibility for the mechanism of the endometriosis, which may provide the theory basis for clinical therpeutic effect.Part ITectological changes of endometriostic lesion in the Uygurs and the Hans in Xinjiang( 一) Observation of the leision for variability between medical therapies and non-medical therapies under the electron microscopeObjective Observation of the leision ultrastructure variability together with the change before and after medical therapies in the Uygurs and the Hans nationality's endometriosis. Methods 41 cases of endometrial cyst of the ovary were observed by JEM-1200 transmission electron microscope which were comfirmed by laparotomy or celoscope operation and pathological examination in department of gynecology , First Affiliated Hospital, Xinjiang Medical University. Among them 7 cases were the Uygurs and 34 were the Hans nationality, all of them were the third or the forth degree of the endometriosis cyst of the ovary. There were 5 cases the endometrium in the site were observed at same time. 30 cases never used medical therapy and 11 cases used medical therapy for three to six months.Result 1, making use of transmission electron microscope, we found that the ultrastructures of the gland cells of ectopic endometrium, which was not trested by medicine, generally resembled that of endometrial gland cells and eutopic endometrium, including endometrial gland, gore in the capillary, more granular reticulum and mitochondrion, phagolysosome increasing while the other cell organs decreasing, hemorrhages and macrophages that phagocytized red cells or hemosiderin incell gaps and so on. 2, Inside walls of the cystiform EMs that treated by medicine, the histological structures were destroyed. The primary changes were the dominent reticular fiber, more fibrocyte, vacuolar degeneration of mitochondrion, The other cell organsand gland components were seldom 3, The ultrastructures of EMs in the Uygurs were the same as that the Hans nationality. Conclusion 1 , The changes of EMs ultrastructures and of eutopic endometrium and ectopic endometrium had identical changes between the Uygurs and the Hans in xinjiang. 2, The cystiform inside walls of EMs, which was not treated by medicine, their ultrastructures resembled eutopic endometrium , both the new hemorrhage's lesions and the old ones could be seen. 3, The main changes of ultrastructures were the fibrocytogenesis in the cells of cystiform inside walls of EMs treated by medicine. (二) Expression of apoptosic genes Fas and c-myc inUygurs and Hans women with endometriosis in xinjiangObjectives: To evaluate Fas and c-myc expression in ectopic endometrium from women of Uygurs and Hans nationality with endometriosis. Method: Fas and c-myc genes expression were examined in eutopic ,ectopic from patients with EMs women and without EMs using immunohistochemical staining. Results: Fas and c-myc immunoreactivity was found predominately in the glandular cells,c-myc expression in ectopic, eutopic endometrium with EM...
