Objective: To observe pathological changes in rat brain and dynamic changes of neutrophil infiltration as well as leukocyte myeloperoxidase activity after focal cerebral ischemia-reperfusion (CIR). ICAM-1, P-selectin and E-selectin, the correlated adhesion molecules mRNA and protein expressional and astrocyte reaction were investingted.. To discuss preliminaryly inflammatory immunological mechanisms of cerebral ischemia-reperfusion injury caused by adhesion molecules and leulocyte, reveal the acupuncture mechanisms in resisting inflammatary injury. Methods: Male Wistar rats were randomly divided into five groups: sham-operation group, model group, non-acupoint group, traditional points selected acupuncture group (Traditional group) and Xingnaokaiqiao acupuncture group (Xingnao group). MCAO model with nylon suture method was set up. The successful model rats are randomly divided into five time points: lhour ischemia-3h, 6h, 12h, 24h, 48h reperfusion respectively(n = 6-13). The acupoints in Xingnao group are "shuigou"(GV26), "neiguan" (PC6) and "sanyinjiao" (SP6), while the acupoints in Traditional group are "quchi"(LIll) and "zusanli"(ST36), while two fixed points under axillary region in non-acupoint group. The rats in three acupuncture groups were stimulated with electro-acupuncture needles. The rats are decapitated at corresponding time point. The indexes were as follows:(1) The improving of neural defective symptoms by estimated by neural behaviors .( 2 ) To observe the dynamic changes of pathology with TTC staining , Hematoxylin-Eosin staining, Nissl's staining and ultrastrusture.(3) The dynamic changes of the activity of myelperoxidase by mensurating OD value in spectrophotometer.(4) The dynamic changes of mRNA expressive position and time of ICAM-1, P-selectin and E-selectin by In situ hybridization methods .(5) The dynamic changes of protein expressive position and time of ICAM-1,P-selectin, E-selectin and GFAP by imunohistochemistry techniques.(6) Image analysis by computer picture analysis system. Results:1. The model of focal cerebral ischemia - reperfusion was made by middle cerebral artery occlusion (MCAO) with surgical nylon suture in rats. The injury area is localized in frontal parietal lobe cortex and striatum observed by TTC staining observation. The infarct area is peaked at 24h reperfusion after 1h ischemia, and the infarct focus is already softened and necrosed at 48h reperfusion time point.2. HE staining result showed: The phenomenon of neutrophil adhension and infiltration was found in cerebral cortex and striatum lh ischemia-6h reperfusion, and became serious and the necrosis center magnified gradually. However, it is minished after acupuncture treatment except non-acupoint treatment (p>0.05) , and the phenomenon is not distinct or the degree alleviated in traditional group and Xingnaokaiqiao group. The improving degree in Xingnao group is better than that in Traditional group (p<0.05).3. The MPO activity became increased in the striatum and cortex lh ischemia -6h reperfusion and increased more post 24h reperfusion, peaked at post 48h reperfusion. It was reduced significantly after acupuncture treatment (p<0.05) except non-acupoint treatment(p>0.05), and the curative effect in Xingnao group was better than that in Traditional group (p<0.05).4. The mRNA expression of ICAM-1, P-selectin and E-selectin began to detect at capillary endothelia cells in cerebral ischemic cortex and corpus striatumn lh ischemia-3h reperfusion and peaked at 6~12h reperfusion, and increased continually during 48h reperfusion by in situ hybridization. They were downregulated significantly treated by acupuncture (p<0.05) except non-acupoint treatment (p>0.05). The downregulated degree in Xingnao group was better than that in Traditional group (p<0.05).5. The outcome by Imunohistochemistry approved that the protein expression of ICAM-1, P-selectin and E-selectin was similar in ischemic area to that of mRNA...
