Objective:Our objective is to study sequential E-selectin and myeloperoxidase expression in cerebral tissue after rat cerebral ischemia-reperfusion,to ascertain the neuroprotective mechanisms of Atorvastatin.Methods:seventy-two healthy male Sprague-Dawley rats(240-260)g were randomly divided into control group,experimental group and Atorvastatin treated group.Each group was divided into subgroups(each group has six animals):4hours group,8hours group,12hours group,24hours group. Sprague-Dawley rat focal cerebral ischemia-reperfusion models were established following the Longa suture method. In the control group, received only anesthesia and vascular separation.Each animal in the intervention group was received6.5mg/kg Atorvastatin daily one week before the operation.To judge the model bases on Zealonga’S scoring and TTC staining results.We detected E-selectin and myeloperoxidase expression in rat brain tissue with immunohistochemical methods.Experimental results were analyzed with the statistic soft package of Spss17.0.Results:E-selectin expression:We found few positive expressions at each time point in the control group (4h,8h,12h and24h). In the experimental group, after cerebral ischemia-reperfusion, the visible positive cells were largely in the cortex and hippocampus. E-selectin was expressed at4h. By12h, expression had reached its peak. At24h, expression return to normal levels. However, in the Atorvastatin treated group, E-selectin expression was significantly lower than in the operation group at all time points (p<0.05).Myeloperoxidase had a similar timing of expression changes to E-selectin, and the levels were significantly lower in the treated group than in the operation group (p<0.05).Conclusion:(1) Brain E-selectin and Myeloperoxidase increased after cerebral ischemia-reperfusion,and changed dynamically with the time of ischemia. E-selectin and Myeloperoxidase are positively correlated. It is Showed that E-selectin and Myeloperoxidase are involved in cerebral ischemia-reperfusion of pathological process.(2) Atorvastatin can decrease E-selectin and Myeloperoxidase expression, atorvastatin may protect against acute cerebral ischemia-reperfusion through anti-inflammatory effects.
