Objective To determine the frequency and type of p16 gene homozygous deletion and mutation of human gastric carcinomas of different periods with the progression of human gastric carcinogenesis, and it relates to clinicopathology, in order to it affords one effective molecule-biologic guideline for early diagnose and early treat of gastric carcinomas Methods PCR (Polymerase Chain Reaction), PCR-SSCP( Single strand conformation polymorphism ) and DNA sequencing were performed on 50 operative tissues, which includes 7 early gastric cancer and 43 advanced gastric cancer. Results 1 . Total mutation frequency of p16 gene was 8%, including 14.29%of early cancer and 6.79%of advanced cancer. , there was not a significant difference between early cancer and advanced cancer (P>0.05) . In addition, p16 gene mutation was not related to site, location, grade of tumor differentiation and lymph node metastases, respectively (P>0.05) . 2. Total deletions frequency of p16 gene was 16%, of which 18.61% was detected in advanced cancer, but no cases in early cancers, there was a significant difference between early cancer and advanced cancer(P<0.05) , and frequency pl6 gene homozygous deletion was related to grade of tumor differentiation and lymph node metastases, respectively(P<0.05) . Conclusion 1 .p16 gene mutation is early event in gastric carcinogenesis , It helps early diagnosis of human gastric carcinoma..2.p16 gene homozygous deletion is late event in gastric carcinogenesis, it might relate to metastases and recurrence of gastric cancer.
