Objective:To explore the relationship and significanceamong the alteration of P16 P53 Rb gene and biological behavior of bladder transitional cell carcinoma(BTCC). Method: 63 cases of P16 P53 Rb expression in BTCC were examined by styeptavidin-peroxidase conjugated(S-P)method; 23 cases of P16 P53 gene deletion and mutation were examined respectively by polymerase chain reaction(PCR) and polymerase chain reaction single-strand conformation polymorphism analysis(PCR-SSCP) in BTCC. ResuIts:(1)The positive expression for P16 P53 Rb were 46.0%(29/63),50.8%(32/63),47.6%(30/63),respectively. All above genes expressions were related to clinic stage(p<0.01);P16 Rb gene expressions were related to the tumor grade(p<0.05); (2)The deletion retio of P16 gene in exon 2 was 17.27%(4/22),the mutation may not be involved in BTCC; The mutation retio of P53 gene in exon 5-8 was 27.27%(6/22),the deletion was not detched. (3)There was the inverse relationship between P16 and P53, P16 and Rb(p<0.05). Conclusions:(1) The results suggested P16 P53 Rb gene play important roles in occurance and development of BTCC,P16 gene major in deletion and P53 major in mutation. (2) The antioncogenes of both Rb(Rb-CyclinDl/CDK4/6-P16)and P53(P53-P21WaF1-Cyclindl/CDK4/6-Rb)pathways lie in relationship possibly.
