Functional Assessment Procedure and Test Methods of Health Food that ChineseSanitary Department has just issued is the method specification of domestic detectionand appraises function food function property. It has found during practice anddetection that the reliability of the data resource was related with the tested number ofanimals, the feeding environment, the individual difference and the detection method.However the reliability of the assessment results was established based on theresearch of particular experiment environment, skilled experiment technology,desirable animal model and accurate statistics analysis. Therefore, this paper hasstudied on function appraisement method of anti-fatigue and anti-hypoxia with fourkinds of different functional liquids as received samples, and has researchedemphatically on the following crucial technologies:(1) Basing on the basic project defined in Functional Assessment Procedure andTest Methods of Health Food, which was issued by Chinese Sanitary Department, theresearch adopted the method combined sport experiment with livelihood index hasbeen adopted, the experiment indexes of little rat loaded swimming , serum ureaexperiment and liver sugar survey were used to assess the anti-fatigue effect offunctional liquid of sampleⅠ. But some factors such as the feeding temperature, watertemperature of swimming experiment and experiment time would impact the time ofloading swimming of mice. The method combined orthogonal test L8(27)has beenapplied to research the effecting factors of experiment time, water temperature ofswimming experiment, feeding temperature and their mutual action in loadedswimming of china medicine function liquidⅠ, the orders of effecting factors of loadingswimming applied by extreme difference (R low) was: Water temperature of swimmingexperiment > experiment time > (water temperature of swimming experiment ×experiment time) > feeding temperature > (experiment time ×feeding temperature).The best combination in the experiment of loaded swimming of china medicinefunction liquidⅠwas: the test lasted for 30days, the water temperature of swimmingexperiment was 25℃and the feeding temperature was 25℃. Therefore, the accuracyof loaded swimming experiment should be increased, or it would result in contingencyof anti-fatigue appraisal results and unavoidably cause the phenomenon of badreappearance. (2) In the light of Functional Assessment Procedure and Test Methods of HealthFood released in 1996 by China health ministry, oxygen lack in normal atmosphere ofmice, which is the item of fundamental test, and the acute anti-hypoxia and the NaNO2poisoning which are added on complementary stipulation 1999, the research focusedon the property of anti-hypoxia of four kinds of medicine fluid Ⅰ~Ⅳ. The result of theresearch shows:Under the condition of oxygen lack in normal atmosphere of mice, theacute anti-hypoxia and the NaNO2 poisoning, they were all masculine when mice weretested by medicine fluid, so it could be conduced that there are some effects ofanti-hypoxia in the medicine fluid. (3) The result of measuring the periphery blood targets of mice shows: differentexperiment time and stay time caused different results in Erythrocyte(RBC),Hemoglobin(HB),The bulk of condensation of the erythrocyte(HCT),Equallyerythrocyte bulk(MCV),Equally the erythrocyte hemoglobin(MCH),Equallyerythrocyte hemoglobin concentration(MCHC).There will be some fruits if the alterablesituation of the periphery blood targets of mice, which tested only 30days or 15days,were used in evaluating their anti-hypoxia. Therefore, in order to get accurate andobjective evaluating results, the periphery blood targets of mice should be syntheticallyanalyzed by different experiment time and stay time. Further more, the research foundout that mathematical model strutted by the relationship between the periphery bloodtargets of mice and stay time, not only decrease experiment times and the amount oftested mice, but also decrease error caused by the difference of individual mouse. Itcan also decrease the fortuitous factors of functional evaluation, and overcomedissimilarity of the appearance of result. (4) The effect of anti-hypoxia and anti-fatigue evaluated comprehensively basedon eight biochemistry which was glutamate (ALT), glutamic oxaloacetic transaminase(AST), lactate dehydrogenate (LDH), creatinine (CREA), alkaline phosphates (AKP),glucose (GLU), urokinase (CK), urea-nitrogen (BUN) in the mice's serum wasmeasured. The research found out that different experiment time and stay timeeffected eight biochemistry properties differently and the alter laws among theproperties were so hard to find that it can't directly judge the effect property ofanti-fatigue or anti-hypoxia. By correlation and analytic hierarchy process, a newmodel that was suit for comprehensive evaluation of anti-hypoxia and anti-fatigue forseveral serum properties was established. It can not only avoid complicated statisticson various related targets that interrupted accurate valuation, but also convenientlyand quickly valuate anti-fatigue and anti-hypoxia properties of the eight biochemistryand avoid failing to analyze caused by the differences of individual sample. In addition,using this model to evaluate the four tested samples, the process of evaluation wasforthright and the result of evaluation was audio-visual and its conclusion was thesame to the former one. (5) Basing on the periphery blood targets of tested mice and serum biochemistryaltogether 23 targets, the research on evaluation of anti-hypoxia and anti-fatigue foundout that when the rich experiment statistics were got, the analyzing process becamecomplicated. If we evaluate the samples that were chosen by experience or by random,the conclusion of the analyzing will be unilateral and the statistics can't be usedsufficiently. If we make use of all the statistics to synthetically evaluation,every targetmust be evaluated and then synthesize the conclusions of all the targets. However, theconclusion of the evaluation will have contrast, and the amount of statistics willincrease. Therefore, in order to avoid the troubles caused by synthetically evaluationand make use of experiment statistics, the dissertation made use of an importantanalyzed method of multivariate statistics ---main composition analysis method, thatwas comprehensive variable and reduce dimension, then the major functional effectindex (main composition) related directly with anti-fatigue and anti-hypoxia effect hasbeen further determined, and according to quantification of functional effect property ofsample a new appraising method has been established basing on main compositionsynthesize. (6)The method of anti-hypoxia and anti-fatigue quartering evaluation byperipheral blood and serum biochemistry property, we found that, could not onlyconquer the trouble of statistic and analysis brought by comprehensive appraisement,but also avoid the discordance of the final comprehensive appraisement result broughtby variety indexes, furthermore it could reduce really animal experiment times,increasing experiment reappearance and accurate. It was a convenient, quick andaccurate evaluation method. The method was not only useful for optimizing usinganimals in selecting "functional divisor"in the anti-fatigue effect of functional liquid ofsampleⅠ,but also fit to any synthetically evaluation research fields in anti-fatigue andanti-hypoxia. (7) The research of optimizing the best dosage and time of anti-fatigue andanti-hypoxia has found: Compared with conventional method, according to thecombination method of orthogonal polynomial mathematics model and geneticalgorithms, the best dosage and time of notable anti-fatigue effect has been optimized.The polynomial mathematics model that reflected the change of hemoglobin contentwith different tested time was used to determine the best time with notable effect ofanti-hypoxia by getting the biggest value of equation. It could optimize accurately, getthe best dosage and time of notable effect, and make the research result moreaccurate and credible and the judge argument more ample. it has conquered...
