Fatigue is becoming the important issues in modern life which can not be ignored, especially the chronic fatigue syndrome (CFS) which comes under a extreme fatigue condition in a long period is a very common sub-health problem in the 21st century. Currently there are some traditional medicines which have anti-fatigue effects, this study focuses on the anti-fatigue pharmacological effects of an extract from traditional Chinese medicine which called Oenanthe Javanica and made a implemented progressively studies about it.We started the research from the anti-hypoxia, anti-exercise fatigue experiments to figure out the efficiency of OJTPA on anti-hypoxia and anti-exercise fatigue. First, the anti-hypoxia research was performed mainly through determining the survival time of mice under a two weeks administration. The anti-exercise fatigue research was conducted first by building up an exercise model with a running machine for mice, then determining the anti-fatigue function parameters such as blood lactate, blood urea nitrogen, hepatic glycogen and exhaustive time, as well as free radicals, oxygen transport capacity of red blood cells. The results obtained that OJTPA can increase the survival time of mice under the hypoxia and low pressure and the exhaust time of mice in the wheel of fatigue instrument, which significantly improves reserves of glycogen, decreases the concentration of blood lactate and blood urea nitrogen, proving that OJTPA has a good efficiency against exercise-induced fatigue, and the mechanism of the anti-fatigue effect may be related to significantly improving the level of free radicals and blood oxygen transport capacity.In addition, we also carried out the anti-deficiency research to evaluate the anti-fatigue effect of OJTPA. Through hydrocortisone we made the anti-deficiency model and fed mice with OJTPA. After two weeks of drug feed, the level of c AMP, cGMP, and hormone level in the serum were determined. The results obtained that OJTPA can significantly improve the level of cAMP, cGMP and hormone level when decreased in deficiency. The mechanism may be connected to the improvement in the status of endocrine, especially the function of hypothalamus-pituitary-adrenal axis (HPA) or gonad axis.In the study of the influence of OJTPA on the status of endocrine, we examined the effect of OJTPA against chronic fatigue syndrome which caused by the stress. We prepared a working model by testing mice swimming in cold water after drug feed. After two weeks, tests of the level of ACTH and CORT for the function indicators of HPA axis,5-HT and DA for the indicators of central fatigue, Ca2+-Mg22-ATP enzyme and pi in the skeletal muscle for the indicators of peripheral fatigue were carried out. The results show that OJTPA can significantly improve the disorder of HPA, and significantly improve the degree of central fatigue and peripheral fatigue.To figure out the effect composition of OJTPA, we tried to study the main compositions and functional groups of OJTPA by methods including infrared spectroscopy, UV spectroscopy and nuclear magnetic resonance. The results show that OJTPA contains some hydroxyl and carboxyl compounds which may have aromatic structures.In order to build up a method for maintaining the efficiency level of OJTPA, we prepared PLA microsphere loading with OJTPA and measured the drug release performance of the microspheres ontained. The results indicate that the microspheres have smooth but porous surfaces and show controlled drug release behavior.
