| Objective: To optimize the technological parameters in extracting antioxidants from pomegranate peels and determine the contents of total phenolics, flavonoids and tannins, as well as their scavenging capacity against various radicals. The hypolipidemic effects of different fractions from pomegranate peel extracts were further investigated and the underlying mechanism was discussed. Some components in the ethyl acetate fraction were separated and characterized.Methods: The concentration of solvent, ratio of solvent volume to pomegranate peelweight and the suitable time were considered as the parameters to optimize the extracting procedure by Response Surface Method (RSM). The crude extract was further separated by ethyl acetate, and the contents of total phenolics, flavonoids, tannins were compared. The antioxidant capacity of various fractions was measured by different methods. The hypolipidemic effect of various fractions was evaluated on hyperlipidemic SD rats. Different chromatographic techniques were used to separate the active components of EtOAc fraction. Mass spectra were applied to characterize the possible structure of each active component.Results:1. The concentration of acetone , the ratio of solvent volume to pomegranate peel weight and the extracting time were the key factors in the extracting procedure,and they were 61.2%, 29.1:1 and 5.85hr, respectively.2.
